標題: 台灣中部地區豬隻寄生蟲相調查及應用臭氧處理豬大腸纖毛蟲囊體和豬蛔 蟲蟲卵之探討

摘要: 自1997年7月至1998年2月,調查台中縣及彰化縣轄內10個養豬場,在每一 飼養欄內逢機取一糞便為材料,共取236欄樣本,所有樣本皆以塗抹法、 浮游法及沈澱法並用D''antoni''s iodine solution染色後檢查。另取台中 、彰化地區屠宰場的144個豬血清,以latex凝集試驗檢查弓蟲( Toxoplasma gondii)抗體反應。亦在台中縣及彰化縣轄內取5個養豬場的 三階段廢水加以檢查。此外,為了從豬糞中純化大腸纖毛蟲( Balantidium coli)囊體以便做進一步的實驗,我們嘗試利用mesh或 percoll來純化,希望找出一種簡便而優良的囊體純化法。再者,我們運 用臭氧來處理本次調查中最結果各種寄生蟲之總感染率為:豬蛔蟲15.6 %(37/236),豬鞭蟲(Trichuris suis)4.6 %(11/236),豬腸節結蟲 (Oesophagostomum dentatum)1.2 %(3/236),縮小包膜絛蟲( Hymenolepis diminuta)0.4 %(1/236),球蟲(coccidia)19.4 %(46/236),大腸纖毛蟲55.5 %(131/236),芽胞阿米巴( Blastocystis sp.)6.7 %(16/236),內阿米巴原蟲(Entamoeba sp. )29.6 %(70/236),體外寄生蟲(ectoparasite eggs)19.9 %(47/236),Rhabditis larvae 0.8 %(2/236)。其中芽胞阿米巴為台 灣豬隻中首次的報告。而且飼養於水泥地面的豬隻所有寄生蟲感在144個 血清檢體中,有28個(19.4 %)呈現弓蟲抗體陽性反應。而在廢水調查方 面,第一階段廢水可檢出豬蛔蟲卵、豬鞭蟲卵、豬球蟲卵囊及外寄生蟲蟲 卵;第二階段廢水只可檢出豬蛔蟲卵及外寄生蟲蟲卵;第三階段廢水則全 無檢出。應用percoll與saline以1:1之比例混合來純化大腸纖毛蟲囊體 ,可獲得良好的純化效果。在25℃下,大腸纖毛蟲囊體經臭氧處理40分鐘 後,約有90 %的囊體已經破損。然而臭氧不能直接破壞豬蛔蟲蟲卵,反可 促使其胚化速率提昇(p<0.01)。
A total of 236 swine fecal samples in central Taiwan were randomly collected to investigate the endoparasitic fauna from July 1997 to February 1998. Direct smear, sugar solution flotation method and sedimentation techniques were employed with D''antoni''s iodine solution stain for examination of parasite ova and oocysts. A total of 114 sera of swine were also collected from slaughterhouses in central Taiwan for detecting antibody to Toxoplasma gondii by latex agglutination test. Wastewater from the three-stepThe prevalence of the following parasites are : A. suum 15.6 %(37/236), Trichuris suis 4.6 %(11/236), Oesophagostomum dentatum 1.2 %(3/236), Hymenolepis diminuta 0.4 %(1/236), coccidia 19.4 %(46/236), B. coli 55.5 %(131/236), Blastocystis sp. 6.7 %(16/236), Entamoeba sp. 29.6 %(70/236), ectoparasite eggs 19.9 %(47/236), Rhabditis larvae 0.8 %(2/236). Among the parasites detected, this is the first report of Blastocystis sp. in swine of Taiwan. Higher prevalences of parasitic infection weOf the 144 sera being checked for T. gondii infection, 28(19.4 %)were found to be positive. A. suum eggs, T. suis eggs, coccidia oocysts and ectoparasite eggs were detected in primary stage treated wastewater; only A. suum eggs and ectoparasite eggs in secondary stage treated wastewater; and no parasite eggs nor oocyst in tertiary stage treated wastewater.Purification of B. coli cysts from swine feces using a mixture of percoll and saline in a ratio of 1:1, was found to give the best result. About 90 % of the B. coli cysts treated with ozone for 40 minutes at 25℃ were destroyed as confirmed by light and scanning electron microscopy. Although ozone could not directly destroy A. suum eggs , it hastened the rate of embryonation of A. suum eggs.
