摘要: 自1995年8月起至1997年3月止,自台灣中部地區共計取樣338隻成年 流浪貓 內臟屠體,分別進行解剖檢查,糞便檢查、橫膈肌 壓片檢查、刮取小腸黏膜鏡 檢及組織切片檢查。本次檢查共計 檢出線蟲類、絛蟲類、吸蟲類和原蟲類四類 。線蟲類包括:貓蛔 蟲35.2%(119/338)、貓鉤蟲32.5%(110/338)、犬心絲蟲1.5%(5/338)、 偽皮囊翼胃蟲0.6%(2/338)和毛細線蟲卵,陽性頭數共計199頭, 感染率58.9%;絛蟲類包括:瓜實絛蟲32.5%(110/338)、貓絛蟲17.8 %(60/338) 和孟松裂頭絛蟲14.5%(49/338)。陽性頭數共計191頭, 感染率56.5%,吸蟲類僅 檢出單睪吸蟲,陽性頭數75頭,感染率21.9% ,原蟲類僅檢出貓球蟲,陽性頭數9 頭,感染率2.7%。橫隔肌壓片檢 查未檢出肌肉旋毛蟲(Trichinella spiralis) 的仔蟲,膽囊中並未 檢出中華肝吸蟲。病理學檢查方面:鉤蟲和偽皮囊翼胃蟲等吸血性寄生蟲 造成黏膜出血,單睪吸蟲和瓜實絛蟲等寄生於腸道絨毛之間、壓迫 周圍組織,偶見黏液分泌增加,幾乎不引起炎症反應,在檢出犬心絲蟲之 貓的肺 臟中檢出血管內皮增生和肌肉層增厚以及嗜酸性球聚集的現 象。單睪吸蟲人工 感染實驗,寄生部位以空腸黏膜為主,感染10日 的回收率較14日為佳。貓蛔蟲的 生化特性分析方面:以隨機增殖聚 合酵素連鎖反應方法進行DNA指紋圖譜分析 ,並以犬蛔蟲和禽類蛔蟲 作為比較。使用逢機合成之十基引子(primer)對各由20 隻來自 不同宿主的雌雄蛔蟲之DNA混和溶液進行RAPD-PCR 反應,由140組引子中 篩選出10種反應較穩定之引子進行分析,每個一引子增殖出1-3個相同之 DNA片段 共得到77個DNA片段,其中24個為共有之特定片段,構 築出貓蛔蟲之指紋圖譜。 使用相同引子對犬蛔蟲和禽類蛔蟲 進行RAPD-PCR反應構築指紋圖譜,相較之下 貓蛔蟲與犬蛔 蟲之間有部份DNA片段相同,另有些DNA片段具有相當明顯的差異, 此點與型態學鑑定上相符。
A total of 338 visceral organs of stray cats in central Taiwan was examined for endoparasitic helminths from August, 1995 to March 1997. Of the 338 cats examined, 285 were infected with endoparasites, with a breakdown of 118 cats being infected with only one species, 97 cats with two, 50 cats with three, and 20 cats with four species of helminths, respectively.Of the 338 cats, 119 were positive for Toxocara cati, 110 for Ancylostoma tubaeforme, 5 for Dirofilaria immitis, 2 for Physaloptera pseudopraeputialis, 110 for Dipylidium caninum, 60 for Taenia taeniaeformis, and 49 for Spirometra erinacei. The prevalence of endoparasites in cats showed seasonal in variation. High incidence of T. cati and D. caninum were observed spring, and A. tubaeforme and T. taeniaeformis in autumn. The prevalence of S. erinacei in cat was high in winter and D. immitis high in summer. Haplorchis taichui artificially infected cats. The major parasitic site was in jejunium mucosa. The retrive rate was below 10%. Pathology examination : found the parasites, gastric and intestine mucosa blooding, mucus sedretion increase, villi around the parasite were atrophy, sometimes eosinophills infiltration in the mucosa or endothelium cell around the parasites. Establishment of DNA fingerprinting in Toxocara cati by RAPD PCR. Ten primers were selected from 180 different ramdon ten-base oligonucleic primers by DNA pools of 20 Toxocara cati both male and female individually. The 10 primers was performed by RAPD-PCR and and a total number of 77 specific products was obtained includ 24 specific products was commonly. Some fragments were same and some different between T. cati and T. canis. This result was the same with the observation of morphology.
