標題: 本省中部地區乳牛乳房炎病原細菌的種類, 抗藥形態, 及乳房鏈球菌之分 子流行病學分類研究
Studies on the distribution of bovine mastitis pathogenes, anti- biotic resistant patterns, and molecular epidemic classification of Streptococcus uberis.

摘要: 本實驗以 API 20 STREP 系統行乳房鏈球菌生化特性的確認與分析,結果 可區分成13種 API 型,出現率依序為 Ⅰ 型(46.1%)、Ⅱ 型(15.8%)、Ⅲ 型(9.2%)、Ⅳ 型(7.9%)、Ⅴ 型(6.6%)、Ⅵ 型(3.9%)、Ⅶ 型(2.6%)、 Ⅷ 型到 ⅩⅢ 型均為 1.3%,其中以 Ⅰ 型(BCN 7443730)出現率最高。 在乳房鏈球菌分離株血清學鑑定方面,本實驗共選取B、C、D、E、G、P 及 U等七種血清型進行檢測,結果顯示,可區分成 E、U、P、D 及 B 等 五種單一血清型,與 D+E、E+P、E+U 及 E+P+U 等四種複合血清型。其 中 E型單一血清型的分離率為 67.1%(51/76),另外有 76.5%(13/17)的牧 場分佈率,為最普遍存在的乳房鏈球菌。若將血清型與生化特性之 API型 結合成 SA 型(Serovar-API combination type)作進一步分析時,結果產 生 21種 SA 型。此外亦對乳房鏈球菌的拒水性(Hydrophobicity) 進行研 究,發現具有堆聚現象(Clump phenomenon)生長特性者,若以Chi- square test 進行統計分析時,顯示出與細胞表面的拒水性有極高的相關 性。在切割酵素指紋圖譜(RFLP)與引子擴合片段指紋圖譜(DAF) 的分析方 面,本實驗以 HindⅢ 之核酸限制切割酵素進行切割片段分析,在 9-23 kb 的區分範圍中,76 株乳房鏈球菌可區分成 50 種 RFLP 型。另一方面 ,利用聚合■連鎖反應( PCR ) 進行引子擴合片段指紋圖譜的分析,在三 段不同的引子中,對於 SA 型的分辨率,分別為引子 A (73.7%)、引子 B (63.2%) 及引子 C (64.5%),而三段引子組合的總分辨率為 92.1%。
Biochemical profils of Streptococcus uberis were analyzed by API 20 STREP system . Totally 13 API patterns were identified during this analysis. The distribution rates of each pattern were as followed:patternⅠ(46.1%), patternⅡ(15.8%), pattern Ⅲ(9.2%) , pattern Ⅳ(7.9%), pattern Ⅴ(6.6%), pattern Ⅵ(3.9%), patternⅦ(2.6%), pattern Ⅷ to ⅩⅢ(1.3% each). Pattern Ⅰ(BCN 7443730) had the highest isolation rate among 13 API patterns. 7 kinds of antisera, including Lancefield type B, C,D,E,G,P and U, were chosen to identify serovars of76 Streptococcus uberis strains. The result of these precipitin test revealed 5 mono- specific serovars(E,U,P,D,B) and 4 patterns of combination sero- vars (D+E,E+P,E+U,E+P+U). Among these versatile serovars, type E was found to have the highest isolation rate (67.1%;51/76) and farm distribution rate (76.5%;13/17). Further analysis by com- bining the result of API type and serovar was able to create 21 SA types(serovar-API combination types) among these 76 strains. An interesting clump phenomenon during bacterial growth was observed among many strains of Strep. uberis. Using salt aggre- gation test and Chi- square analysis, We discovered a significant relationship between this clump phenomenon and cell surface hy- drophobicity. With regard to restriction fragment length polymorphisms- (RFLP) analysis, 50 RFLP patterns, which adopted Hind Ⅲ as restriction enzyme and distinguished the fragment length poly- morphisms in the range of 9∼23 kb, were exhibited. We also con- ducted a DNA amplification fingerprinting (DAF) investigation. The descrimination rates for 76 strains by using 3 different primers were as followed : primer A(73.7%), primer B(63.2%), primer C(64.5%), and primer A+ B+C(92.1%).
