摘要: 本研究利用CAS屠宰廠之豬肝作為試驗原料,利用三種不同酵素 Papain、Alcalase,Protease N水解12小時,每小時蒐集水解液後測定pH值、可溶性蛋白與胜肽含量變化。再選用胜肽含量最多之水解液作為最佳水解條件之後進行動物試驗。結果顯示,三種酵素皆有水解豬肝之能力,而Alcalase (A2)與Protease N (Pn2)在兩小時水解後可使胜肽含量達到最高,Papain水解之產物則需4小時(P4)才具相同之效果。以此時間點作為最佳水解條件進行冷凍乾燥後,進行體外抗氧化試驗與CCK分泌試驗後,可知三種水解物皆有良好之抗氧化效果與CCK分泌能力。每日灌食經高脂飼糧誘導之大鼠後進行血液分析,三種酵素水解物皆可降低風險因子(Risk factor, LDL/HDL)。P4水解物與羅氏鮮處理組可顯著降低血漿中瘦體素之含量( p < 0.05),雖然利用P4和A2之水解液可顯著降低大鼠體重( p < 0.05),但無法顯著( p < 0.05)降低腎周脂肪與副睪脂肪之重量,可知水解液對於抑制臟器脂肪形成並無較明顯之功效。採食量方面,灌食三種水解物顯著降低其採食量( p < 0.05),而對照組顯著地有較高之採食量( p <0.05)。綜觀上述,推測水解液藉由採食量下降,以降低體重,其中P4與A2之水解液對於體重之抑制有較好之效果,且可降低心血管疾病之風險,對於水解液抑制體重增加的機制,將做更深入的研究,以開發具有減肥效果之活性胜肽。
In the current study, we utilized porcine livers as the materials collected from CAS slaughter factory for the by-product to examine the bioactivities of its hydrolysates. Three various enzymes, Papain, Alcalase, Protease N were used to hydrolysis the livers for 12 hours. Hourly collected hydrolysates were measured the pH value, soluble protein and peptide contents, and antioxidative activity. The maximal peptide production of the tested hydrolysates was chosen for following bioactivity studies. All of the 3 enzymes exhibited the ability to hydrolyze porcine liver. Alcalase(A2) and Protease N(P4) had the highest peptides production after 2 hour hydrolysis, whereas Papain (P4) needed four hours to reach the same yield. The hydrolysates with highest peptide contents were fed the rats by gastric tubing (500mg/ml) in combination with high fat diet. All of the 3 hydrolysates could decreases the risk factors including LDL-C/ HDL-C. P4 and A2 hydrolysates decreased body weight significantly (p < 0.05) but the decreased body weight were not associated with a consistent decrease in the visceral fat. Both Orlistat, a well-know anti-obese drug and P4, significantly (p < 0.05) suppressed plasma leptin levels in the rats with high fat diet. All of the 3 hydrolysates decreases food intake significantly when compared to the control (p < 0.05). In conclusion, the hydrolysates, P4 and A2, can decrease the body weight by suppressing food intake in association with a lower risk factor. We then expect to isolate the bioactivity peptides and investigate their mechanisms for body weight loss function in the future studies.
