摘要: 本試驗使用紫苞舌蘭品種Spathoglottis plicata 及其兩個變異種粉花苞舌蘭Spathoglottis plicata‘RS01’與白花苞舌蘭Spathoglottis plicata‘RS02’為試驗材料,利用苞舌蘭成熟植株之側芽、花梗節及花芽進行微體繁殖。首先針對改善培植體微生物污染的問題進行探討,接著探討不同細胞分裂素或不同培養添加物對誘導芽體形成與增殖之影響,建立有效的芽體再生路徑,最後探討IBA對苞舌蘭瓶苗發根及生長之效用。 紫苞舌蘭在前處理對抑制微生物污染的試驗中,三種花色的紫苞舌蘭植株只要於栽培期間時施用免賴得(Benomyl)即可明顯改善培植體微生物污染問題,其中又以使用免賴得配合兩週斷水處理有最佳的效果。此外,側芽、花梗節及花芽三種培植體在所有處理組及對照組的內生菌污染百分率表現上,皆以花梗節污染率最高,而花芽的污染情形最少。 不同培植體中,只有側芽培植體能順利誘導出芽體。花梗節培植體不論培養於任何培養基之下,其培植體均產生嚴重褐化,在任何處理下皆無法誘導出任何芽體。花芽培植體雖然同樣於任何處理下皆未能誘導出任何芽體,但其褐化情形較不嚴重,存活的花芽都能繼續成長並伴隨子房膨大。Spathoglottis plicata、Spathoglottis plicata‘RS01’與Spathoglottis plicata‘RS02’的側芽培養於添加8.88 µM BA的培養基中效果較佳,8週後每個培植體分別產生1.89 、2.44及3個芽體,芽體形成率分別為90%、90%及60%。增殖倍率的部分,三者皆以添加8.88 µM BA的培養基增殖倍率最高,4次繼代平均增殖倍率為分別為2.09 、2.26及3.01倍。 發根試驗中,不論是藉由BA或Kinetin所誘導出來的芽體,於IBA的培養下,三種花色的紫苞舌蘭小苗都會發根,發根率皆為100%,培養時IBA濃度建議使用7.38 ~ 9.84 µM,對於小苗的植株發育、根部形成與發育,以及根部活性而言都有最佳效果。
