摘要: 本研究為探討高溫及化學藥劑處理對四季蔥瓶苗去病毒之影響,先以ELISA方法篩選‘蘭陽一號’及‘桃園三號’兩四季蔥品種之罹病株,切取帶三枚葉鞘之莖頂(含基盤)做為培植體,於瓶苗階段進行高溫及化學藥劑處理,以比較兩處理對二品種四季蔥分蔥潛隱病毒去病毒(Shallot latent virus, SLV)之效果,並進行無病毒四季蔥瓶苗之增殖處理,以期達到健康種苗生產之目的。 在高溫處理方面,帶分蔥潛隱病毒之培植體以35℃高溫處理3-4天,可獲得達50%的去病率。去病毒效果隨著處理時間的延長或處理溫度的提高而增加,最佳效果可達到80-100%,在兩四季蔥品種上都有呈現相同的趨勢。 另觀察高溫處理對園藝性狀、葉綠素、可溶性糖、澱粉、總可溶性蛋白質及脯胺酸等含量結果顯示,兩品種經高溫處理後鮮重變化並不顯著。株高、葉綠素及可溶性糖隨著處理溫度增加而降低,澱粉則是隨處理溫度增加而增加。總可溶性蛋白質含量皆以38℃處理的含量最高。脯胺酸含量以40℃處理下含量最高,‘蘭陽一號’及‘桃園三號’ 大約比對照組高2及2.36倍。 在化學藥劑處理方面,兩品種以25 mg/l Ribavirin藥劑處理兩週後,即可完全排除SLV,且在延長培養期間,仍可繼續維持無病毒感染的狀態。但隨著處理濃度之增加及處理時間的延長,培植體鮮重、株高、葉片數及葉綠素的含量都有呈現顯著下降的趨勢。培植體以Dithiouracil藥劑處理者,不論處理的時間及處理濃度為何,去病毒效果都較差。但隨著處理濃度的增加及處理時間的延長,鮮重、株高、葉片數及葉綠素的含量都顯著的優於以Ribavirin藥劑處理。 在芽體增殖方面,以含有5 mg/l TDZ及1mg/l NAA+2 mg/l BA隻培養基培養,對於培植體地上部的生長,具有顯著的促進效果。在發根數方面以MS-free之對照組及5mg/l 2ip及5mg/l Kinetin之處理,能提高發根數。 本研究結果建議利用38℃高溫處理一週或25 mg/l Ribavirin藥劑處理兩週,移至MS+1mg/l NAA+2 mg/l BA增殖培養基培養一個月,再移到不含任何植物生長調節劑的MS基本培養基中,做為培植體之發根誘導,經培養一個月後即可達出瓶栽培之大小。出瓶馴化後可得無SLV病毒之健康種苗。
The objectives of this study were to evaluate the effects of thermotherapy and chemotherapy on Shallot latent virus-elimination in green onion (Allium fistulosm L.). The shoot tips with three leaf sheathes were dissected from Shallot latent virus-infected (SLV- infected) two green onion cultivars ‘Lan Yang No.1' and ‘Tao Yuan No.3'. Subsequently, the regeneration of virus-free plantlets was also tested for the production of healthy seedlings in vitro. Results of thermotherapy revealed a 50 % reduction in virus-infected plants after being treated at 35℃ for 3-4 days. The percentage of virus free plantlets increased as temperature increased and treatment duration prolonged. The best result of 80-100 % virus elimination was obtained at 38℃ with treatment duration of 7 days. In addition, when temperature rose, starch content in plantlets was increased. But there were adverse effects on plant height, chlorophyll and solubility sugar contents. Total soluble protein contents was the highest in thermotherapy at 38℃. The praline content in ‘Lan Yang No.1' and ‘Tao Yuan No.3' treated at 40℃ were 2 and 2.36 times higher than that of control, respeatively. Results of chemotherapy revealed that SLV was eliminated completely by the medium with 25 mg/l of Ribavirin and cultivated for 2 weeks. However, there were adverse effects on fresh weight, height, number of leaf and chlorophyll content of explant as the chemical concentration and cultural period increased. The chemotherapy effect of Dithiouracil was not satisfied at any concentration, although it showed beneficial effect on plant height, fresh weight, leaf number and chlorophyll contents. Results of explant proliferation showed that the plantlets cultured in MS medium containing 5 mg/l TDZ or 1 mg/l NAA and 2 mg/l BA could be promoted for shoot regeneration significantly. MS-free and MS medium with 5 mg/l 2ip or 5 mg/l Kinetin could promote the root induction of green onions. It is suggested that the green onion materials can be treated at 38℃ for one week or cultured in medium containing 25 mg/l Ribavirin for two week to eliminate SLV-virus. After the thermotherapy or chemotherapy, the plantlets are transfer to MS medium containing 1 mg/l NAA and 2 mg/l BA for proliferation. After one month, the plantlet are further transferred to plant grow regulator-free MS medium for another one month for rooting. After acclimatization, the SLV-free health seedling cam be derived.
