摘要: 本研究利用無刺麒麟花‘粉仙子’經秋水仙素誘變而得到的多倍體誘變株13株(MC01~MC13)與大花麒麟Euphorbia × lomi ‘Red Giant’(RG)、‘Sonia’(S)、‘Sri Aumphorn’(SA)、‘Supo Roek’(SR)、‘Udom Sub’(US)以及陳(2012)育出之雜交種‘SAg02’進行雜交,期能育成無刺的大花麒麟。經Brewbaker and Kwack (1963)花粉培養基培養,各誘變株的花粉培養在含糖15~20%時花粉發芽率較高。再經不同溫度培養,各誘變株的花粉,在25~30℃下花粉發芽率較高,且發芽率皆高於19.7%以上,顯示誘變株可以做為育種親本。在授粉試驗中,‘粉仙子’誘變株之自交結果率可達32.8%,共獲得37株自交後代。在與E. × lomi雜交,除了與‘Supo Roek’或‘Red Giant’雜交分別獲得雜交後代319株或1株外,其餘雜交組合皆無法產生後代。在‘Supo Roek’ × MC01~MC13雜交所得到的後代再自交、回交與互交中,只有SR × MC01、SR × MC10或SR × MC11再與MC11回交時能夠得到4株回交後代外,其餘組合都未得到後代。將雜交組合SAg02 × MC01~13授粉後17天之未熟種子培養,其總發育率可達29.8%。至2013年5月止共計有5株出瓶。‘Red Giant’ × MC01~13於授粉後13、17天之未熟種子培養,以17天成熟度者發育較佳,其總發育率可達34.4%且共有4株出瓶。將‘Udom Sub’ × MC01~13於授粉後13、15、17天後之未熟種子培養,15天成熟度者發育較佳,總發育率可達14.3%且已有2株出瓶。雜交後代在刺的表現上,結果並未得到完全無刺的個體。所有雜交組合產生的後代皆屬於多刺型,但是在刺的硬度卻有程度上的差異。在‘Supo Roek’ × MC01~13的255株雜交後代中,其中有175株為軟刺,80株為硬刺。‘Red Giant’ × MC06只獲得1株雜交後代,且屬於硬刺。
The plants of Euphorbia milii ‘Pink Fairy’ mutants (MC01~MC13) by colchicine were crossed with E. × lomi ‘Red Giant’(RG), ‘Sonia’(S), ‘Sri Aumphorn’(SA), ‘Supo Roek’(SR), ‘Udom Sub’(US), or ‘SAg02’(Chen, 2012) for creating thornless E. × lomi. The suitable sucrose concentration in Brewbaker and Kwack (1963) medium for pollen germination in vitro of E. milii ‘Pink Fairy’ MC01~13 mutants was at 15~20%. The suitable temperature for pollen culture of E. milii ‘Pink Fairy’ MC01~13 mutants was at 25~35℃, and pollen germination rate of all mutants was above 19.7%. In self-crossing of mutants of E. milii ‘Pink Fairy’ MC01~13, the fruitset rate were 32.8%, and got 37 offsprings. In all crosses with E. × lomi cultivars, only when ‘Supo Roek’ or ‘Red Giant’ crossed as maternal parent was successful. And 319 or 1 offsprings developed, respectively. In the cultured of immature seeds, the successful rate was at 29.8%, when immature seeds of SAg02 × MC01~13 when cultured at 17 days. Until May 2013, 5 offsprings developed. When immature seeds of ‘Red Giant’ × MC01~13 were cultured at 17 DAP, the successful rate was 34.4%, 5 offsprings developed. As well as the successful rate of culturing ‘Udom Sub’ × MC01~13 immature seeds was at 14.3%, and 2 offsprings developed. Thornless individuals did not be created in this research, and all crosses produce offsprings are spiny, but there are different level of thorn hardness. In ‘Supo Roek’ × MC01~13 255 hybrids, there are 175 plants with soft spines and 80 plants with hard spines. There is one with hard spines from ‘Red Giant’ × MC06.
