摘要: 扇形文心蘭在植物分類上隸屬於文心蘭亞族,具有短幼年期,可在試管內開花的特性,適合進行組織培養誘變育種。扇形文心蘭的類原球體經秋水仙鹼或氨磺靈處理,皆能誘導出多倍體植株,以0.1% 秋水仙鹼振盪培養4天,有最高的擬多倍體誘導率77.1%,經0.01% 氨磺靈固體培養3週,擬多倍體誘導率為64.3%。扇形文心蘭多倍體成熟植株的植株厚度、最大葉片厚度和最大根直徑皆較二倍體植株厚,並可使用做為早期篩選標的性狀。多倍體植株染色體觀察,結果顯示同時有整倍體株、非整倍體株及嵌合體株,多倍體株間染色體數有差異,同一個根尖樣品的染色體數也不同。多倍體株花色呈深黃色,花型呈現差異,花萼及側花瓣較為圓寬,柱頭與花粉粒皆增大,葉片單位面積的葉綠素a及葉綠素b含量都較二倍體株高。從誘變株中挑選出具觀賞或研究價值之單株供日後使用。
The plant classification of Erycina pusilla was belonging to Oncidiinae, had a short juvenility and in vitro flowering characteristics, suitable for mutation breeding by tissue culture. PLBs (protocorm like bodies) of Erycina pusilla through colchicine or oryzalin treatments can induce to regenerate polyploid plants. Inducing liguid medium supplied with 0.1% colchicine shaking for 4 days had the highest rate of 77.1% of polyploid induction, and 0.01 % oryzalin treated for three weeks had 64.3% of polyploid induction. The plant thickness, maximum blade thickness and maximum root diameter of the mature polyploids were higher than diploids, and could be used as the subject of early screening traits. Chromosome examination of polyploid plants showed presence of euploidy, aneuploidy and chimerias. Polyploid chromosome numbers were different between plants, and chromosome numbers of root tips from the same sample were also different. In addition, polyploid flowers had saturate yellow in color, more round and width in sepals and lateral petals, and increased in lengths and widths of stigma and pollen size. The chlorophyll a and chlorophyll b per unit area of polyploids are higher than diploids. Mutants with ornamental or research value could be selected for future use.
