摘要: 為尋找誘引東方果實蠅(Bactrocera dorsalis (Hendel))雌蟲的有效成分,本研究結合嗅覺電生理技術與氣相層析儀化學成分分析,篩選出植物揮發性氣味中對東方果實蠅雌性成蟲可感受成分。前人研究指出爪哇鳳果(Garcinia dulcis Kurz.)葉片萃取物具有極佳的誘雌(>90%)和誘引產卵效果,因此採用活性碳通氣裝置蒐集爪哇鳳果葉片的揮發性成分,再以氣相層析質譜儀(Gas chromatography-Mass spectrometry, GC-MS)分析鑑定其化學成分,結果發現葉片的揮發性成分主要為倍半萜烯類(Sesquiterpene)化合物中的cubebene, copaene, caryophyllene, germacrene D, α-caryophyllene, elemene, cadine-3,9-diene。氣相層析-觸角電圖(Gas chromatography-electroantennographic detection, GC-EAD)同時分離爪哇鳳果葉片的揮發性成分與偵測觸角對感受成分的反應,可再進一步篩選出誘引雌成蟲的可能成分。試驗結果發現爪哇鳳果葉片萃取物在分離後8-11分鐘內出現GC和EAG (Electroantennogram) 相對應的峰值,再以購得的商品化有效誘引成分cubebene, copaene, caryophyllene做GC-EAD反應的比對,發現雌蟲對caryophyllene有明顯的EAG反應。此外,在30 cm3白色壓克力飼養箱內放置裝有不同劑量caryophyllene誘餌的透明寶特瓶進行2-14日齡的100對混合飼養的東方果實蠅雌、雄蟲及100隻分開飼養的雌蟲的誘引效果測試發現,溶於hexane的100㎕caryophyllene對上午捕獲再放回(morning returns)的第9日齡混合飼養的雌蟲的平均誘引率最高為72%,而促使雌蟲產卵試驗方面,則以溶於hexane的100㎕caryophyllene促使上午捕獲再放回的第10日齡混合飼養的雌蟲在裝有caryophyllene之透明寶特瓶內產最多卵,1小時內平均產50粒卵。由上述結果可證實在30 cm³白色壓克力飼養箱中,透明寶特瓶內放置的caryophyllene誘餌具有誘雌與促使產卵的效果。
In order to identify the effective components for attracting female oriental fruit fly, Bactrocera dorsalis, this research employed electroantennogram (EAG) technique and gas chromatography (GC) to investigate the volatile components of Garcinia dulcis leaves. Previous studies have shown that extracts of the leaves of G. dulcis was very attractive (>90%) to the female oriental fruit fly and numerous eggs were laid by the attracted females. Therefore, aeration assembly was employed to collect the leaf volatilize components of G. dulcis. With chemical analysis by GC-MS, the collected leaf volatile components were Sesquiterpene compounds, including cubebene, copaene, caryophyllene, germacrene D, α-caryophyllene, elemene, and cadine-3,9-diene. GC-EAD was then applied to identify the effective components. The leaf volatile components of G. dulcis in 8-11 minutes appeared the peaks after the separation which GC and EAG correspond. With the commercially available compounds of cubebene, copaene and caryophyllene, the mixed breeding of female has obvious EAG response to caryophyllene, but not cubebene and copaene. In addition, the attractive test uses the different dose caryophyllene has carried on to 100 pairs of 2~14-day-old mixed breeding of females and males and 100 separated breeding of females, the result showed that 100㎕caryophyllene dissolves in hexane catches 72% of the morning returns 9th age mixed breeding of female. Behavioural observation also showed that 100㎕caryophyllene dissolves in hexane elicited the 10-day-old mixed breeding of female to dump more eggs than other ages. These results confirm that the caryophyllene in 30 cm³ cage is the most effective component in the leaf extract of G. dulcis attracting the female oriental fruit fly.
