dc.description.abstract本研究之目的在於調查進口木材和本土林木中長喙殼菌類真菌的種類,建立正確之鑑定技術。此類真菌除了有性態屬於Ophiostoma屬外,並有四種極其不同之無性形態(歸在不完全菌之Leptographium、Pesotum、Sporothrix、Hyalorhinocladiella),實驗中結合傳統形態、配對試驗與現代分生技術,以達到正確鑑定之目的。結果共鑑定出9種長喙殼菌類真菌,除O. quercus為已紀錄種外,包含4個新紀錄種,分別為O. floccosum、O. pluriannulatum、L. bistatum、L. sinoprocerum,及4個新種,分別為L. floricolonia、L. sparsum、L. tristatum、P. hyalina。將所收集的菌株與基因庫中相似種類進行DNA序列比對,並建構親緣關係圖譜,探討形態特性與親緣之關係。選擇O. quercus為模式進行配對實驗,建立最佳之配對方式及配對測試菌株,未來可以此模式應用到其他長喙殼菌類真菌之鑑定上。zh_TW
dc.description.abstractThe aim of this study is to survey ophiostomatoid fungi from imported and indigenous wood in Taiwan. Four anamorphic genera, Leptographium, Pesotum, Sporothrix, and Hyalorhinocladiella are associated with the teleomorphic genus Ophiostoma. Species identifications were carried out by using morphology, mating compatibility test, and DNA sequences comparison. Adequate approaches of identifying these fungi were therefore obtained. Nine ophiostomatoid fungi were recognized. Except for O. quercus, which has been recorded in Taiwan, there are 4 new records, including O. floccosum, O. pluriannulatum, L. bistatum, and L. sinoprocerum, and 4 new species, including L. floricolonia, L. sparsum, L. tristatum, and P. hyalina. Phylogenetic analyses were conducted by using sequences collected in this study and those of relevant species deposited in GenBank. Morphologically characters were linked to phylogenetic relationship. Isolates of O. quercus were used in the trials to establish the best method of mating compatibility test. It is expected that the established model of mating compatibility test can be applied to other ophiostomatoid fungi.en_US
