摘要: 由 Xanthomonas 屬引起之茄科細菌性斑點病,為茄科植物重要之細菌性病害。自 2001年至 2010 年由臺灣番茄主要栽培區所分離之細菌性斑點病菌,大部分皆屬於 X. perforans。本研究探討市售番茄品種對X. perforans之感受性,及X. perforans在番茄葉表之群集情形。測試九種市售番茄栽培品種對於 X. perforans 之感受性,結果顯示供試之番茄栽培品種皆對 X. perforans 感病, 可在葉片產生典型之穿孔病徵,其中小姑娘品種感受性最低,而 TMB-582 與慧嬌品種感受性最高。測試X. perforans 菌株對農友 301 番茄品種引起之病害嚴重程度,結果顯示 XTN169 菌株造成之罹病度最高。在 X. perforans 對 7 種化學藥劑之感受性測試中,顯示所有供試之菌株對多保鏈黴素 (Thiophenate-methyl + Streptomycin) 的感受性最高,而供試之 8 個菌株中有 6 個菌株對含銅混合劑已有抗藥性產生。測試 Pseudomonas putida YLFP4、YLFP14、YLFP16 與 YLFP18 菌株對供試之 4 個 X. perforans 菌株之抑制作用,顯示在 PDA 培養基上皆有明顯的抑制能力。為瞭解 X. perforans 在番茄葉片之群集情形,本研究利用具綠色螢光蛋白之 pBREGFPexp 質體,以電轉法導入 X. perforans XTN169 菌株,獲得穩定表現螢光蛋白之衍生菌株 XTN169EG。將具綠色螢光蛋白之 XTN169EG 噴佈於番茄農友 301葉片,利用雷射共軛焦顯微鏡觀察 XTN169EG 在葉表增殖群集之情形,結果顯示接種後維持高濕環境所形成之群集較低濕時高。探討 P. putida YLFP14與 YLFP16 對 XTN169EG 在番茄葉片上群集之影響,顯示 YLFP14或YLFP16 可降低 XTN169EG 在番茄葉表之群集。測試 X. perforans 在不同番茄品種葉表之族群量,顯示在感受性最低之小姑娘葉片上之族群量低於感受性高之品種。將P. putida YLFP14 或 YLFP16 (108 CFU/mL) 與 X. perforans XTN169 (106 CFU/mL) 混合後接種於番茄農友301 葉表,可降低細菌性斑點病之發生。
Bacterial spot of solanaceous plants caused by strains of Xanthomonas spp., is an important disease of solanaceous plants. Strains of Xanthomonas associated with bacterial spot of tomato were isolated from fields since 2001 in Taiwan. Most of these strains were identified as X. perforans. In order to study the susceptibility of commercial tomato cultivars to strains of X. perforans and their colonization on the tomato leaves surface, nine commercial tomato cultivars were used to test the susceptibility of tomato cultivars to strains of X. perforans. The results showed that strains of X. perforans can cause shot-hole symptoms on all tested cultivars. The disease severity results showed that the cv. KY Chica is the most tolerant, while the cv. V20148 and Savvy are the most susceptible. The disease severity caused by X. perforans strains on tomato cv. Farmers 301 was conducted in this study. The disease severity caused by X. perforans strain XTN169 on tomato cv. Farmers 301 is the highest. The sensitivity of X. perforans strains to various agrichemicals was examined. The results showed that all X. perforans strains were sensitive to Thiophenate-methyl + Streptomycin. Six out of eight strains of X. perforans were copper compounds resistant. The antagonistic bacteria Pseudomonas putida YLFP4, YLFP14, YLFP16 and YLFP18 significantly inhibited the growth of X. perforans on potato dextrose agar (PDA). To visualize the colonization of X. perforans on the tomato leaves surface, enhance green fluorescent protein expressing vector pBREGFPexp was introduced into X. perforans XTN169 by electroporation, resulting in a stable derivative XTN169EG. The strain XTN169EG was sprayed on the leaves of tomato cv. Farmers 301, and the cells of X. perforans XTN169EG colonized on the leaves of tomato cv. Farmers 301 was examined by confocal laser scanning microscopy. The colonization of X. perforans XTN169EG was increased on leaves at high humidity. Mixed inoculation of P. putida YLFP14 and YLFP16 with XTN169EG , the colonization of XTN169EG on the tomato leaves surface was reduced. Three different tomato cultivars were used to examine the colonization of X. perforans XTN169EG. The results showed that population of X. perforans on cv. KY Chica was less than the susceptible cv. V20148 and Savvy. Co-inoculation of P. putida YLFP14 or YLFP16 in 108 CFU/mL suspension with X. perforans XTN169 in 106 CFU/mL on the leaves of tomato cv. Farmers 301 can reduced the disease severity of bacterial spot .
