標題: 大豆栽培種和野生種自交與種間雜交胚及胚乳發育之研究

摘要: 以栽培種大豆(G. max, cv.KS10)與 Glycine亞屬之野生種G. tomen- tella(To12)、G. dolichocarpa(Do3)、 G. tabacina(Ta2, Ta18)、 G. latifolia(La2)、 G. canescens(Ca1)、G. falcata(Fa1)等7種 8系 統為材料,進行栽培種與野生種之種間雜交, 調查正、反雜交的成功率, 雜交莢長, 並利用石蠟切片觀察不同物種大豆之自交及種間雜交後胚及胚 乳之發育,行組織學之觀察,結果如下: 1.以KS10為母本, 野生種大豆為父 本, 5天內不落莢之正交成功率為 9.44% (309/3271);反交 5天內不落莢 之成功率為 3.32%(21/633); 正交組合中,除KS10 x Fa1延至14左右天落 莢(莢長7mm),其餘各組合可延至20天左右才落莢(莢長10∼27mm),但未得 雜交種子。 2.大豆不同物種自交胚、胚乳切片結果, 除胚乳同屬核型胚 乳外,胚的發育期相當不一致,發育最快者為Ta18,4天胚為圓球期,8天心型 期, 10天已發育達子葉期;其次為La2, 3天為原胚,4天圓球型期,6天心型 期, 12天達子葉期;次為Ta2及Ca1,7天為圓球型期,Ta2 9天,Ca1 11天為 心型期, 13天皆達子葉期;再次為KS10及To12,2天屬原胚,5∼6天圓球型 期,9天心型期, 14天為魚雷期及17天發育至子葉期胚;最慢為Do3, 4天為 圓球型期, 10天為心型期, 18天發育至子葉期。 3.種間雜交胚、胚乳切 片結果,正交: 所有組合, 胚乳均較胚提前敗育, 如 KS10 x To12 在11天 時胚乳不正常分裂, 18天胚發育達子葉期,但胚組織鬆散,有敗育現象; KS10 x Do3 16天胚乳開始敗育,24天胚則仍停滯在心型期;KS10 x Ta2 16 天胚乳組織呈現鬆散, 24 天胚囊內組織已敗育; KS10 x La2 8天胚 乳開始敗育,22天圓球型期胚及胚乳均已敗育;KS10 x Ca1胚在20天為魚 雷期,胚乳已出現敗育;KS10 x Fa1 13天時胚乳即出現敗育,胚則仍停滯 在8天時之圓球型期。反交;To12 x KS10 12天胚乳呈不正常現象,24天時 魚雷期胚才敗育;Do3 x KS10 16天胚乳開始敗育,21天胚珠已萎縮;Ta2 x KS10 23天圓球型期胚及胚囊敗育萎縮。
Interspecific reciprocal crosses were made between G. max (KS10) and G. tomentella (To12), G. dolichocarpa (Do3), G. tabacina (Ta2, Ta18), G. latifolia (La2), G. canescens (Ca1), G. falcata (Fa1) of subgenus Glycine respectively. The successful rates of reciprocal crosses and hybrid pods size were investi- gated. Further, the developmental characteristics of embryo and endosperm of selfed and interspecific cross in Glycine species using with paraffin section were also observed.
