摘要: 在藥物經皮輸送系統發展中,皮膚表面的角質層為藥物經皮滲透最大的阻礙,為了降低藥物分子傳遞的阻礙和提高經皮吸收的效率,常利用物理性、化學性促進方法來提高。本研究目的為評估不同分子量幾丁聚醣與幾丁聚醣濃度促進經皮滲透的能力。實驗中選擇間-苯二酚作為模型藥物並以溫感性聚合物作為藥物載體,將中分子量幾丁聚醣(MC)和低分子量幾丁聚醣(LC)作為促滲劑並製備成不同凝膠配方,並以無添加促滲劑的配方為控制組。實驗使用裝置垂直式Franz Diffusion Cell(FDC)進行體外實驗研究,並藉由數據與模型擬合有效滲透係數。實驗結果顯示,當幾丁聚醣分子量增加會減少藥物的滲透係數。在MC/Poloxamer與LC/Poloxamer配方中,實驗結果顯示,滲透係數與幾丁聚醣濃度呈現非線性關係,分別在濃度0.005g/ml與0.0075g/ml時會出現最佳的滲透係數。
The stratum corneum layer is viewed as the most significant permeation resistance in transdermal drug delivery process. Physical and chemical enhancements are commonly used to reduce the permeation barrier for drug molecular and thus increase the efficiency of transdermal absorption. The purpose of the research is to evaluate the transdermal drug delivery enhancement capacity of chitosan with a various combinations of molecular weights and concentrations. Resorcinol and thermosensitive copolymers were chosen as model drug and dug carrier. Different gels were prepared with Medium molecular weight chitosan (MC), Low molecular weight chitosan (LC), respectively as enhancer, and the control was chosen to be with no enhancer. In vitro permeation studies were carried out using Franz-diffusion cells and the analytical model was employed to fit the effective permeation coefficient with experiment data. The result showed that the drug permeation coefficient decreases as the molecular weight of chitosan increases. The permeability was found to have no positive non negative correlation with the concentration of chitosan. The local maximum of permeabilities were appeared at 0.005g/ml and 0.0075g/ml for MC/Poloxamer and LC/Poloxamer respectively.
