標題: 國產基因改造木瓜回交果實之毒理安全性評估

摘要: 本實驗室於96及97年度分別完成單抗木瓜輪點病毒轉基因木瓜18-2-4及16-0-1及雙抗基因改造木瓜TPY10-4之基因毒理和動物毒理安全性評估,結果顯示,單抗基因改造木瓜之生果及熟果均不具基因毒性和動物毒性,其食用安全性與一般木瓜相同。98年度「國產雙抗基因改造木瓜對大鼠90天動物毒理及腸內菌相安全性評估」,初步結果顯示國產雙抗基因改造木瓜生熟果均不影響腸內有害及有益菌相。本計畫進一步配合國內已進入田間試驗之基因改造木瓜,針對木瓜輪點病毒及畸葉嵌紋病毒單抗18-2-4與單抗親本非基改台農2號回交後(編號22-10)及單抗親本非基改泰國株木瓜回交後(編號823)、及回交後編號22-10與823雜交商業品種F1果實,分別進行基因毒理及動物毒理試驗,國產基因改造上市前食用安全性評估。有關毒理試驗係依據衛生署公告之基因改造產品及臨床前毒理安全性試驗,並符合美國環境保護署(EPA)及OECD之毒理試驗規範進行評估。測試基因毒理試驗,包括微生物基因突變分析、細胞基因毒性測試,動物口服急毒性、28天餵食毒性等試驗,將有助於進一步瞭解基因改造木瓜經由回交後果實之食用安全性,作為國產基因改造上市前食用安全性評估。
During 96 and 97 years, we have conducted the safety assessments for whole foods of genetic modified plant foods of mono (18-2-4 and 16-0-1) and double resistant (TPY10-4) for Papaya ringspot virus (PRSV) and Papaya leaf-distortion mosaic virus (PLDMV) transgenic papaya in Taiwan. Results revealed that whole foods of genetic modified plant foods have no genetic and animal toxicities bewtwen the parent fruits on these studies. For the intestinal bacterial flora cultivation, they also showed no effect on the harmfull or probiotics in the intestines in rats. The objective of this project is conducetd the safety evaluation of the hybrid fruits, included mono-resistance transgenic papaya lines (18-2-4) hybrid with the non-GM control line of Tainon-2 (code no. 22-10), or non-GM Thailand control line (code no. 823) and the other one is hybrid with 22-10 and 823. The test guidelines are following the tests of Department of Health and also meet the test guidelines of USEPA and OECD for the genetically modified foods. For genetic toxicity, three approaches will be conducted including In vitro gene mutation in bacteria (Ames test) and chromosomal aberration test with mammalian cells in culture In vivo micronuclear assay. Furthermore, animal toxicity will be evaluated on the acute oral toxicity and 28 days feeding test. Data from this study will provide usefull safety evaluation for the hybrid of genetic modified plant foods in Taiwan.
