摘要: 在本研究室之先前研究中,由鳳仙花 (Impatiens balsamina L.) 分離之2-Methoxy-1,4-naphthoquinone (MeONQ),具高度抗幽門螺旋桿菌活性。感染幽門螺旋桿菌與胃癌有高度之關聯性,因此本研究接續先前研究,第一部分探討MeONQ對胃腺癌細胞之抑制作用及其機轉,第二部份探討對幽門螺旋桿菌誘發免疫反應之影響。 MeONQ對MKN45胃腺癌細胞呈劑量及時間依存型之抑制活性,50%抑制濃度為24.01 ± 0.38 μM。25~200 μM造成MKN45細胞停滯於S及G2/M期,低劑量 (25~50 μM) 造成輕/中度細胞凋亡。MeONQ具高度壞死MKN45細胞能力,25~200 μM處理劑量之細胞壞死比率為對照組之196~756%。50~200 μM處理造成粒線體跨膜電位明顯下降,為對照組之11~39%。25~200 μM處理致使MKN45細胞於6~10 h間產生高量超氧陰離子 (為對照組之168~336%)。50 μM劑量使MKN45細胞caspase-3蛋白質表現量下降;12.5~200 μM劑量使Bcl-2及Bax蛋白質表現量增加、12.5~50 μM劑量使細胞質之cytochrome c蛋白質表現增加,但使粒線體之cytochrome c蛋白質表現下降。MeONQ對MKN45細胞主要作用機轉為其於細胞內產生高量ROS,啟動Bcl-2路徑,生成更多ROS,產生氧化爆炸,造成細胞壞死;或經Bcl-2路徑,cytochrome c由粒線體釋出,活化caspase途徑造成細胞凋亡。 MeONQ對幽門螺旋桿菌誘發胃癌免疫反應之影響方面,MeONQ使感染幽門螺旋桿菌MKN45細胞之IL-8生成量增加 (為對照組之為302~418%)、ROS (過氧化氫及超氧陰離子含量分別為對照組之85~91%、52~67%) 生成量及COX-2表現量下降,IκBα蛋白質表現量增加。推測MeONQ對幽門螺旋桿菌誘發MKN45細胞免疫反應,係經由NF-κB路徑,影響IL-8、ROS及COX-2等發炎因子蛋白質表現及生成量。
