摘要: 去鹼基核酸(AP sites)為最常見的內源性DNA損害之一,其形成原因為化學去鹼基以及氧化壓力等原因。許多內源性或外源性之藥物,對於哺乳類動物細胞可能使其產生AP sites。此研究之主要目的為探討人類乳癌細胞、乳癌患者及經化學治療之乳癌患者其體內白血球之AP sites背景值。去鹼基核酸大多源自於DNA分子之鹼基修復、醣損害及醛基損害,DNA分子上之氧化損害所造成之AP sites是利用醛基反應探針所偵測總核苷酸而得。結果表明,在人類乳腺癌細胞中MCF-7、T47D與MDA-MB-231中,其AP sites背景值分別約為7.3、9.5與17.8 (每一百萬個核苷酸) 。為了檢驗這些細胞株之AP sites來源,我們將DNA與腐胺反應使其切除DNA之3''端點位置。經腐胺切除實驗結果表明,在乳腺癌細胞中為5''端切除之AP sites約含有65% 。 近一步研究顯示,乳癌化療患者其AP sites背景值約為111 (每一百萬個核苷酸),相較於無使用化學治療之乳癌患者約上升2倍。
