摘要: 本驗試驗採用採自種子園的八個半親家系的實生苗作為試材,每隔一個月進行一批繁殖,研究杉木微體繁殖力的「家系效應」與「批次效應」。 結果顯示,第二批中的家系間產芽量無顯著不同(第20週時,P=0.1392),發根量無顯著不同(P=0.1914),萌芽量亦無顯著的不同(P=0.1914)。而發根率、萌芽率是有顯著不同的(P值分別為0.069與0.008)。在第三批中,家系間的產芽量無顯著不同(第20週時,P=0.9232),發根率無顯著不同(P=0.491),萌芽率無顯著不同(P=0.295),發根量無顯著不同(P=0.7980),萌芽量無顯著不同(P=0.8737)。不同批間的產芽量不同(在增殖階段P=0.0732,在發根階段P=0.0594)、發根率有顯著不同(P=0.019),發根量不同(P=0.1966)、萌芽率不同(P=0.080)、萌芽量不同(P=0.2009)。 在不同批的比較中,有些項目雖未達一般統計的顯著水準,但還是粗略的認為不同批培養間是有差別的。然而本試驗只有兩批,所以批與批間雖有不同,但不能因此就認為「批次效應」有很大的影響力。而且全部上述結論都還有許多疑點待澄清,包括:操作人手不同可能導致的變異、使用的數量化參數不恰當以及實生苗樣本數不夠多。
Seedlings collected from eight half-sib families in the seed-orchard were used as experiment material. We conduct a batch of micropropagation every month to investigate “family effect” and “seedling age effect” of the production potential in china fir micropropagation . The result shows that in the second batch, the difference of bud number among families are not significant(at the 20th. week, P=0.1392). The difference of root number among families are not significant(P=0.4743). The difference of sprout number among families are not significant(P=0.1914) . But root rate and sprout rate show significant difference among families in this batch(P=0.069 and 0.008, repectively). Meanwhile, in the third batch, the difference of bud number among families is not significant(at the 20th. week, P=0.9232). The difference of root number among families is not significant(P=0.7980). The difference of sprout number among families is not significant(P=0.8737). The difference of root rate among families is not significant(P=0.491). The difference of sprout rate among families is not significant(P=0.295). On the other hands, the difference of bud number between batches is significant(in the multiplication stage P=0.0732, in the rooting stage P=0.0594). The difference of root rate between batches is significant(P=0.019). The difference of root number between batches is significant(0.1966). The difference of sprout rate between batches is significant(P=0.080). The difference of sprout number between batches is significant(P=0.2009). In the tests between batches, some results do not achieve traditional statistical significant level but we roughly conclude that there are differences between batches. In fact, we have only two batches in our experiment, so we can not conclude that “batch effect” has key influence to micropropagation of china fir. Besides, there are quite a few questions need to be clarified. First, different workers handling different batches may cause variations between batches. Second, quantitative parameters used in the experiment may not be appropriate. Third, seedlings materials used in the experiment may not be enough.
