摘要: 雞傳染性鼻炎菌(Avibacterium paragallinarum)為引起雞隻傳染性可利查病(Infectious coryza, IC)之病原菌,此疾病又被稱為傳染性鼻炎,發病的雞隻常可見的臨床症狀包含眼窩下竇腫脹、發出囉音、打噴嚏等症狀,若是感染產蛋雞可造成死亡或是產蛋率下降,進一步導致經濟損失。雞傳染性鼻炎菌為格蘭氏陰性菌,格蘭氏陰性菌有多種致病因子,大多與細胞壁組成相關,其中之一為磷酸化膽鹼(Phosphorylcholine, ChoP)。磷酸化膽鹼是重要的致病因子之一,存在於細菌的最外層,可對黏膜系統發生作用,表現磷酸化膽鹼的菌株可增強細菌黏附於黏膜細胞並且可降低菌體對抗菌胜肽的敏感度,經基因相位改變(Phase variation)可改變脂寡醣之構造和其抗原變異性使的疫苗保護力不盡理想。經由全基因序列分析發現雞傳染性鼻炎菌含有一段序列licABCD與Haemophilius influenzae有高度相似性,此段序列與磷酸化膽鹼修飾於脂寡醣調節相關。其中licA基因含有一段四核甘酸重複序列5’-CAAT-3’,經由菌體增殖複製時Slipped strane mispairing(SSM)可改變重複序列數目,以進一步影響調控磷酸化膽鹼修飾與否。西方墨點法(Western blot)結果證實經由licA基因相位變異(phase variation)可調控磷酸化膽鹼修飾於脂寡醣之與否。抗菌胜肽(antimicrobial peptide)被發現普遍存在多種生物體,屬於先天性免疫的一環,可毒殺入侵的病原體,抗菌胜肽殺菌試驗顯示,有磷酸化膽鹼修飾的菌株可幫助菌體降低對於抗菌胜肽fowlcidin-1的敏感度,以提高菌體之存活率。本研究為第一個發表雞傳染性鼻炎菌LPS表面有磷酸化膽鹼修飾之報導,並發現雞傳染性鼻炎菌之基因序列licA帶有一段四核甘酸組成之重複序列磷酸化膽鹼的修飾能幫助雞傳染性鼻炎菌躲避宿主抗菌胜肽的攻擊,此發現對於瞭解雞傳染性鼻炎菌之致病機轉有重要助益,並以本實驗結果判定有磷酸化膽鹼修飾之菌株較適合當作將來疫苗研究之發展菌株。
