摘要: The root fragments and leaves discs of 'Seven beauty' and 'viva beauty' cultivars of sponge gourds were used to study the changes in protein synthesis under various flooding duration. Protein synthesis was mwasured by 35S-methionine incorporation and analyzed by two-dimensional sodium lauryl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) and fluorography. The labeled polypeptides profiles of non-flooding control and treatments of different flooding duration (1/2, 1, 3 and 5 days) were quite similar. There were 11, 13, 20, and 15 of induced polypeptides in leaves of 'seven beauty' sponge gourds, 2,4,3, and 2 of induced polypeptides in roots, and 11, 8 ,16, and 16 of repressed polypeptides in leaves after 1/2, 1, 3 and 5 days of flooding, respectively. There were 14, 10, 13, and 12 of induced polypeptides in leaves of 'viva beauty' sponge gourd, 3, 5, 4 and 4 ofinduced polypeptides in roots, and 4, 12, 11, and 12 of repressed polypeptides in leaves, after 1/2, 1, 3, and 5 days of flooding, respectively. The changes of those polypeptides among treatment can be classified into five group. Group 1 polypeptides were enhanced or repressed only at certain flooding period. Group 2 polypeptides were remained enhancement or repression for 1 day of flooding. group 3 polypeptides were unique to flooding stress. Group 4 polypeptides were decreased but not repressed completely under flooding. Group 5 polypeptides were unique to aerobic environment.
本試驗利用活體內放射性35S-甲硫胺酸標記絲瓜根部及葉片的蛋白質,並以二次元電擁分析新合成蛋白質的螢光顯像圖譜,探討耐淹水及不耐淹水之絲瓜品種在不同淹水時間,其根部及葉片蛋白質合成之變化。試驗結果顯示'七美'絲括(耐淹水)葉片及根部在淹水1/2、1、3及5天之處理,其新誘導合成之polypeptides在葉片各有11、13、20及15個,而跟布各有2、4、3及2個;合成被抑制之polypeptides在葉片各有11、8、16及16個。'景美'絲瓜(不耐淹水)的葉片在淹水1/2、1、3及5天的處理,其polypeptides被誘導合成的各有14、10、13即12個,根部則各有3、5、4及4個;合成被抑制之polypeptides在葉片各有4、12、11及12個。不同淹水天數處理期間個別polypeptides心誘導合成或被抑制的消長,約可分為五類:1.單獨再某一淹水時間合成被促進或被抑制者;2.被促進或抑制合成之polypeptides,在淹水環境可持續一天者;3.淹水處理特有者,即淹水處理期間合成軍備促進者;4.在淹水期間合成量減少,但沒有完全被抑制者;5.氧環境特有者,即淹水期間合成均被抑制者。
