摘要: 仔豬於出生時腸道菌群不穩定與離乳時受到之緊迫,再加上集約化飼養造成之環境髒亂,使仔豬易受到病原菌之侵害而產生下痢與生長遲緩等問題,而本土中藥草具有抗菌活性,添加益生菌能轉換其有效成分以增強抗菌性,因此本研究之目的為添加益生菌與中藥草(黃芩與梔子)組合於母豬及仔豬飼糧對仔豬生長、改善下痢及腸道菌群之影響。試驗分為兩個部份,第一部份為評估中藥草水萃液(200 mg/ml與50 mg/ml)、益生菌(Lactobacillus acidophilus LAP5、L. casei L21、L. fermentum P2、Pediococcus acidilactici LS)及中藥草水萃液與益生菌共培養對Escherichia coli、Salmonella choleraesuis、S. enterica subsp. enterica、S. choleraesuis、S. typhimurium及厭氧菌Clostridium perfringens之抑菌效果測試。結果顯示共培養之處理較益生菌或中藥草水萃液(200mg/ml Chinese herbs water extracts, CHW)對兼性厭氧病原菌之抑菌效果為佳,而中藥草水萃液(50 mg/ml)對厭氧菌之抑制效果較共培養處理佳。第二部份分為三個動物試驗,試驗分為四個處理組,分別為對照組(空白飼料)、抗生素組(Flavomycin)、飼糧添加0.25%益生菌與中藥草組合(添加0.1% 109CFU/g益生菌與0.1%中藥草組合)及飼糧添加0.125%益生菌與中藥草組合(添加0.05% 109CFU/g益生菌與0.05%中藥草組合)。試驗一,以16頭懷孕母豬分為四組,試驗為期6週,每組四重複,每重複4頭。結果顯示,母豬飼糧添加0.25%益生菌與中藥草組合組能提高母豬哺乳期採食量(P<0.05),並抑制哺乳仔豬腸桿菌、大腸桿菌群與沙門氏菌之趨勢(P<0.1),降低哺乳仔豬下痢情形;試驗二,以320隻離乳仔豬分為四組,為期4週之試驗,每組四重複,每重複80頭。結果顯示,添加益生菌與中藥草組合於飼糧不影響仔豬生長性狀(P>0.05),而飼糧添加0.25%益生菌與中藥草組合組顯著維持小腸絨毛高度及絨毛高/腺窩深之比值(P<0.01),並降低第二週腸桿菌及大腸桿菌群菌數之趨勢(P<0.1),顯著改善第三與四週下痢情形(P<0.05);試驗三,以80隻弱小離乳仔豬分成4組,進行相同處理組之試驗,每組20頭。結果顯示飼糧添加0.125%益生菌與中藥草組合組能提高弱小仔豬0-14天體增重(P<0.05),與顯著抑制第一週沙門氏菌數,在第三週有較低的腸桿菌、大腸桿菌群及沙門氏菌數之趨勢(P<0.1),顯著降低第二至四週糞便指數與下痢發生率(P<0.05),降低腸道pH值與糞氨濃度之趨勢(P<0.1)。綜合上述,益生菌與中藥草組合添加於母豬及離乳仔豬飼糧具有抑制腸桿菌、大腸桿菌群及沙門氏菌等致病菌生長,維持腸道健康,進而改善哺乳仔豬、離乳仔豬及弱小仔豬生長及下痢之情形。
Born piglets intestinal flora unstable or piglets weaned often stuffed a variety of stress, and environmental pollution caused by intensive farming, so that piglets susceptible to pathogens result in diarrhea, growth retardation and other issues. Native Chinese herbal medicine has antibacterial activity, adding probiotics can convert its active ingredients to enhance the antibacterial. Therefore, the purpose of this study was to investigate the effect of probiotics and Chinese herbs (Scutellariae radix and Gardenia fructus) combination in the diet on the growth and intestinal character of piglets. The study is divided into two parts. Firstly, the antibacterial effect of Chinese herbs water extracts (CHW, 200 mg/ml or 50 mg/ml), probiotics, probiotics culture with CHW on E. coli, S. choleraesuis, S. enterica subsp. enterica, S. choleraesuis, S. typhimurium (facultative anaerobes) and Clostridium perfringens (anaerobe) was evaluated. The results showed that the antibacterial effect of probiotics culture with CHW (200 mg/ml) on facultative anaerobic pathogens was better than probiotics and CHW; however, the antibacterial effect of anaerobic bacterium to CHW (50 mg/ml) is better than probiotics and probiotics culture with CHW. Secondly, divided into three animal experiments, and the experiment was divided into four groups: control group (blank feed), antibiotic group (Flavomycin), the diet was supplemented with 0.25% probiotics and Chinese herbs combinations (supplemented with 0.1% of 109 CFU/g probiotics plus 0.1% Chinese herbs combination), and the diet was supplemented with 0.125% probiotics and Chinese herbs combinations (supplemented with 0.05% of 109 CFU/g probiotics plus 0.05% Chinese herbs combination). In experiment 1, 16 pregnant sows were tested, four replicates per group, each repetition of 4 sows. The results showed that 0.25% group could increase feed intake during sow lactation (P <0.05) and inhibit Enterobacteria, coliform and Salmonella in suckling piglets (P <0.1), and reduce diarrhea in suckling piglets. In experiment 2, 320 piglets were tested, four replicates per group, each repetition of 80 piglets. The results showed that the combination of probiotics and Chinese herbs in the diet did not affect the growth character of piglets, and the 0.25% group significantly increased villus height and villus height/crypt depth (P <0.01) and decreased the number of Enterobacteria and coliform in the second week (P <0.1), significantly improved the third and fourth diarrhea (P <0.05). In experiment 3, 80 weak weanling piglets were randomly assigned to receive the same treatment, with 20 weak piglets in each group. The results showed that 0.125% group could increase the weight gain of the weak weaned piglets from 0 to 14 days (P <0.05), and significantly inhibited the number of Salmonella in the first week and the lower level of Enterobacteria, coliform and Salmonella in the third week (P < 0.1), significantly reduced the fecal index and the incidence of diarrhea (P <0.05), improve intestinal pH and fecal ammonia concentration (P <0.1). In conclusion, the combination of probiotics and Chinese herbs added to sows and weanling piglets diet can inhibit the growth of pathogens and maintain intestinal health, thus improving the growth and diarrhea of suckling piglets, weanling piglets and weak piglets.
