骨折癒合可分為三個時期,包括血腫炎症期、骨折修復期及骨痂重塑期。而基質金屬蛋白酵素-9(matrix metalloproteinase-9, MMP-9)在骨折部位中所扮演的是細胞外基質分解酵素的角色。骨折後的血腫炎症反應時,MMP-9活性表現增加以降解細胞外基質,有助於炎症細胞的移動;在骨痂重塑時,MMP-9活性表現的增加有助於骨痂的重吸收。本研究利用組織病理學並配合受質凝膠電泳偵測不同時期未復位骨折骨痂組織中MMP-9活性表現的差異。實驗樣本來源為中興大學獸醫教學醫院小動物外科之骨折門診病例,均以外科手術取得未復位骨折之骨痂組織,共31例。經細胞外蛋白酵素萃取,以500倍稀釋後進行受質凝膠電泳。結果顯示,血腫炎症期及骨痂重塑期之骨痂組織,在92kD處呈現明顯的明膠溶解條帶,而骨折修復期之骨痂組織則不會出現此一條帶。經統計分析後,MMP-9活性表現與骨折後至復位前的持續時間間隔有相關性(p<0.05),但與病畜體重、年齡、骨折斷面以及斷肢部位並無相關性。持續追蹤骨折病例,依據骨折後至復位前的持續時間間隔分組與術後恢復情況進行統計分析,結果顯示,術後恢復情況與骨折後至復位前的持續時間間隔並無相關性。

Bone healing can be divided into three phases, including the inflammation phase, the reparative phase and remodeling phase. Matrix metalloproteinase-9 (MMP-9) lyses the extracellular matrix in a fracture state. In inflammation phase, the high expression of MMP-9 is contributive to the migration of the inflammation cells resulted from degrading extracellular matrix. In addition, the high expression of MMP-9 is conducive to remodeling of bony callus during remodeling phase. This study used zymography to detect the difference of MMP-9 activity in bony callus at those distinct phases before fracture-reduction and also coordinated with the classification of histopathology. Zymography samples were diluted to 1/500 after extracted from extracellular proteinase. Thirty-one samples of bony callus before fracture-reduction were obtained by surgical intervention in Veterinary Teaching Hospital of National Chung-Hsing University. The result of this studying reveals that the zymography displayed a clear band on 92KD in samples of the bony callus of the inflammation phase and the remodeling phase, but not the reparative phase. Through the statistical analysis, it appeared a high relation (p < .05) between the expression of MMP-9 and the duration from fracture to reduction. However, the weight, age, fracture shape and fracture location did not reveal high relation with the expression of MMP-9. Continues to trace the bone fracture cases, we found that the recovery of post-surgery and the duration from fracture to reduction were not related.
