本研究之目的在於嘗試開發檢測福馬林之高效液相層析儀 (High-performance liquid chromatography, HPLC) 技術,並進行福馬林使用於鱒魚及鱘龍魚的安全性及其在肝臟及肌肉的殘留研究。實驗結果顯示本研究所建立的HPLC檢測技術: C18管柱 (5C18, 4.6 I.D. × 250 mm) ,移動相為乙睛 (Acetonitrile):去離子水 (DDW) (20:80 (v/v);pH 7) 、流速1 mL/min,紫外光偵測器之波長設定為360 nm。最低檢測極限可達0.01 ppm。福馬林在鱒魚及鱘龍魚之肝臟及肌肉之回收率分別為 (96.5%和97%) 及 (94%和95%)。毒性試驗結果顯示,隨著福馬林的濃度降低,死亡率也跟著逐漸降低,且兩者之間呈現一種線性關係。於50 ppm福馬林藥浴之下,可發現鱘龍魚及鱒魚的鰓部皆有輕微上皮細胞剝離、剝落及變性等現象,但96小時後魚隻之鰓部開始有復原之狀況。殘留試驗結果顯示25 ppm浸泡組之鱘龍魚肝臟中的甲醛殘留量,肝臟於48小時後即低於最低檢測極限,而肌肉部分則無甲醛殘留。50 ppm藥浴組魚隻之肝臟於96小時後即低於檢測極限,肌肉部分也是無甲醛殘留。鱒魚25 ppm藥浴浸泡組之肝臟於96小時後即低於檢測極限,肌肉則於8小時後即低於檢測極限,而50 ppm藥浴浸泡組魚隻之肝臟於240小時後即低於檢測極限,肌肉則於第24小時後低於檢測極限。綜合結果顯示,福馬林於25及50 ppm濃度下用於鱘龍魚及鱒魚是安全的,使用後240小時皆低於檢測極限,建議其藥物停藥期在鱘龍魚及鱒魚分別為8天及20天。

The aim of this study is to develop of the formalin detection technique by High-performance liquid chromatography (HPLC) and determine the residual concentration of formalin in muscle and liver of sturgeon and rainbow trout. Our results showed, our developed HPLC method set with C18 coloume, acetonitrile: DDW (20 : 80) as a mobile phase (pH 7) , flow rate 1 mL/min, and the UV-detector detects the max peak at wavelength 360 nm. Our results suggested that the median lethal concentration (LC50) lowered progressively as the duration of exposure increased. The toxicity curves became almost asymptotic after treatment 96 h. The 96h-LC50 of sturgeon and rainbow trout was 182.4 and 179 ppm at 19℃, respectively. After 50 ppm formalin treatment, fishes has been found to cause gill lamellar epithelia separation, hypertrophy, and necrosis under the histopathology examination. However, the gills had recovered after 96 h. The time of undetectable residue amount of formalin in the liver and muscle tissue of the sturgeon at 25 and 50 ppm was (48 hr and 96 hr) and (0 hr and 0 hr) , respectively. At the same way, the time of undetectable residue amount of formalin in the liver and muscle tissue of the rainbow trout at 25 and 50 ppm was (96 hr and 240 hr) and (8 hr and 24 hr) , respectively. Thus, to use low concentration of formalin in the edible tissues of cold water fish is safe and recommend. We recommended withdrawal period for sturgeon and rainbow trout were 8 and 20 days.
