墨綠凹鼻魨 (Tetraodon nigroviridis)為棲息於河口的廣鹽性魚類,可適應於海水及淡水,先前的研究已從墨綠凹鼻魨中發現FXYD家族中的一員,命名為pFXYD9,並發現在長期馴化至淡、海水環境時,鰓上pFXYD9在mRNA及蛋白表現量為淡水高於海水,和鰓上Na+, K+-ATPase(NKA)活性及蛋白質含量趨勢相反,顯示pFXYD9 以抑制的方式調控NKA活性。本研究將長期馴養於海水(高張)環境中的魚直接轉移至淡水(低張),透過偵測鰓上pFXYD9基因及蛋白量的變化,搭配NKA蛋白量及活性的趨勢,以及血液滲透壓、氯離子濃度和肌肉含水量等滲透壓調節相關的生理反應,探討滲透壓調節過程中pFXYD9蛋白質所扮演的角色,來了解墨綠凹鼻魨面臨低張環境時的生理機制。
生理方面,在轉移12小時後血液滲透壓和血液中氯離子濃度出現顯著性低點,鈉離子濃度也在24小時明顯下降,隨著體內離子濃度及滲透壓降低,鰓上pFXYD9蛋白質含量和NKA活性開始顯著性變化,NKA活性48小時明顯下降、在96小時顯著升高,血液滲透壓及離子濃度在96小時回升, NKA活性也藉由pFXYD9蛋白質的影響在轉移後168小時顯著下降,由此顯示墨綠凹鼻魨在適應低張環境時,藉由鰓上pFXYD9蛋白質以帶動性影響NKA活性的模式,協助墨綠凹鼻魨調控滲透壓來達成體內恆定。
