本試驗利用PCR-RFLP(polymerase chain reaction-restriction fragment length polymorphism)方式分析豬PNAS-4、裂解刺激因子次單位1(cleavage stimulation factor subunit 1, CSTF1)、肌動蛋白α2(actin alpha 2, ACTA2)、脂蛋白元M(apolipoprotein M, APOM)、胰島素受體基質1(insulin receptor substrate 1, IRS-1)、脂肪重量與肥胖相關基因(fat mass and obesity-associated , FTO)基因之基因型,目的為建立豬隻SNP選拔育種之依據。以軟體設計專一性引子對,經由PCR反應增殖標的DNA片段,以異丙醇沉澱純化PCR產物,再利用PCR-RFLP進行基因型鑑定,最後以變方分析進行基因型與性狀間之比對。分析台灣地區藍瑞斯、杜洛克、約克夏、藍瑞斯 × 杜洛克、藍瑞斯 ×約克夏 × 杜洛克及黑毛豬等豬種各基因之基因型與交替基因頻率分析,並將上述基因之基因型與性狀資料相互比對,發現PNAS-4基因:杜洛克品種AC型個體之背脂厚度顯著高於AA型者。CSTF1基因:約克夏品種CC型個體之背脂厚度顯著高於CT及TT型個體,藍瑞斯 × 杜洛克品種CT型個體之平均日增重與屠體長顯著高於TT型者。ACTA2基因:藍瑞斯品種CT型個體之平均日增重顯著高於CC型個體,藍瑞斯 × 杜洛克品種TT型個體之背脂厚度與屠體長表現顯著優於CC型個體,藍瑞斯 × 約克夏 × 杜洛克品種TT型個體之背脂厚度顯著低於CC及CT型個體,黑毛豬則以TT型個體之肋骨數顯著最少。APOM基因:杜洛克品種CC型個體之背脂厚度顯著高於GG型者,黑毛豬則以GG型個體之肋骨對數顯著較多。IRS-1基因:杜洛克品種AA型個體之背脂厚度顯著高於CC及CA型個體。FTO基因:約克夏品種GG型個體之平均日增重顯著高於CC型個體,藍瑞斯 × 杜洛克品種CC型個體之平均日增重顯著高於GG與GC基因型;CC型個體之肋骨對數則顯著低於CG與GG型個體。上述結果有助於豬隻育種選拔之參考。
