”達豐”結球萵苣,於25 ℃下種子發芽為98 ﹪,種子經35 ℃7天熱抑制處理後,再移至25 ℃下,會降低51 ﹪之發芽率,長期於35 ℃下發芽率為0 ﹪。光照處理可增加種子2 ﹪之發芽率及縮短0.3天之平均發芽天數。
利用—1.1 Mpa PEG8000滲調處理3天及10 mM GA3+ 10 ppm Ethrel + 100μM BA處理後,萵苣種子於35 ℃下平均有87.3 ﹪之發芽率。經35 ℃ 7天熱抑制處理後,以荷爾蒙處理,有23.3 ﹪之發芽率。種子經2天40 ℃100 ﹪RH老化處理後,老化處理後之種子於25 ℃有68.9 ﹪發芽率。在35 ℃下,經滲調或荷爾蒙處理有32 ﹪之發芽率。
經老化處理及高溫浸潤下,會導致種子內35 ﹪總過氧化物及脂質過氧化物之含量增加,20 ﹪SOD、CAT,35 ﹪POD、10 ﹪GST之過氧化物清除酵素活性降低,及30 ﹪GR、GSSG、GSH之抗氧化酵素及抗氧化物含量下降。經滲調及荷爾蒙處理後,可改善種子在35 ℃下之過氧化作用。種子於35 ℃較25 ℃下會降低15 ﹪之蛋白質含量、15 ﹪之可溶性糖、25 ﹪澱粉分解酵素及二氧化碳、乙烯釋出量為零。老化種子於35 ℃下,會降低種子內39 ﹪蛋白質之含量。滲調及荷爾蒙處理可提高種子於35 ℃下增加39 ﹪之可溶性蛋白質35 ﹪可溶性糖含量及1倍之α-β澱粉分解酵素之活性,其呼吸率及乙烯釋出量分別由零增至26 μl / gFW/ hr之及20 nl / gFW/ hr。
種子於25 ℃下,經滲調處理後可增加Mr. 66.2 KD、35.6 KD、32.4 KD、27.2 KD之蛋白質含量。於35 ℃下,滲調增加Mr. 66.2 KD、35.6 KD、32.4 KD、27.2 KD含量之蛋白質,於Mr. 33.8 KD、31 KD處有出現新合成之蛋白質。

Summary
The germination percentage of "MESA659" lettuce seeds was 98 ﹪at 25 ℃. After seeds imbibed at 35 ℃ for 7 days, the seeds had 51 ﹪germination at 25 ℃. The seeds can not germination at 35 ℃. The seeds had 87.6 ﹪germination at 35 ℃ when seeds were primed by -1.1 Mpa PEG8000 for 3 days and 10 mM GA3+ 10 ppm Ethrel + 100μM BA treatments. Seeds were increased 2 ﹪germination and the mean days shorten 0.3 days after light treatment. The germination of artificially aged seeds were 68.9 ﹪at 25 ℃. Priming and hormone treatments could enhance 32 ﹪germination of artificially aged seeds at 35 ℃.
The amount of total peroxide and malondialdehyde were increased, the activity of superoxide dismutase, catalase, peroxidase, glutathione reductase, and glutathione-S-transferase were derceased. The amount of glutathione oxidized form, glutathione reduced form, and glutathione were derceased in artificially aged seeds and seeds imbibed at high temperature. Priming and hormone treatments could improve the antioxidation of seeds imbibed at high temperature. The activity of α-amylase, β-amylase, and the content of soluble carbohydrate of seeds imbibed at 35 ℃decrease 26 ﹪, 24 ﹪, and 15 ﹪, respectively. The amount of soluble protein of seeds imbibed at 35 ℃ decrease 15 ﹪compared to seeds at 25 ℃, moreover the protein amount of aged seeds decrease 39 ﹪. The amount of soluble protein, soluble carbohydrate, CO2,and ethylene were increased in primed seeds and seeds treated hormone at 25 ℃ and 35 ℃.
The amount of Mr. 66.2, 35.6, 32.4, and 27.2 KD protein increased on primed seeds imbibed at 25 ℃ and 35 ℃. Primed seeds imbibed at 35 ℃ appeared Mr. 33.8 KD and Mr. 31 KD.
