本研究以二種芭菲爾鞋蘭,觀察蒴果與種子之發育,Paph. purpuratum授
粉後8個月,種子已部分轉黃,9個月後種子轉為黃褐色,蒴果於11個月時
轉黃色,以掃描式電子顯微鏡觀察,授粉後 1個月,胞原細胞分裂生成一
突起長軸,2-4個月為大孢子發育階段,至5個月種子與種皮形成而脫離胎
座,之後種子逐漸成熟,種子外形增大,胚亦明顯膨大,至11個月時才可
發芽。而Paph. philippinense授粉後105天時,種子為金褐色,120天即4
個月)時全為暗褐色,蒴果於110天時轉褐色,120天後會裂開,以掃描式
電子顯微鏡觀察,授粉後 1-2個月,胞原細胞分裂生成一突起長軸,且進
行大孢子發育階段, 3個月後種子與種皮形成而脫離胎座,逐漸成熟外形
增大, 4個月時才會發芽。三種芭菲爾鞋蘭之種子,播於不同培養基成分
及不同溫度之環境條件下, Paph. purpuratum以MS之培養基且25℃下,
發芽達19.5﹪;Paph. philippinense以RE培養基配方,在25℃下發芽最
好,發芽率達85.5﹪; Paph. sukhakulii亦以RE培養基配方,在20℃下
發芽最好,發芽率達27.1﹪;但當發芽後,原球體由黑暗中轉至25℃並光
照下,原球體有轉綠的情形。Paph. sukhakulii在25℃下發芽者,移至25
℃並光照下,原球體在全部不同的培養基都有增加的趨勢,以1/2MS(A)
之培養基最好,20℃下發芽者移至25℃並光照下,只以1/2MS(A)之培養
基較好,且有陸續發芽之現象;Paph. philippinense原球體照光後,只
以RE+MS(B)+MS(C)+MS(D)之培養基,原球體有稍微增加的趨勢
;Paph. purpuratum仍以 MS之全量配方培養基較好。移植後小苗生長情
形,Paph. sukhakulii以1/2MS(A)+20g/l蔗糖濃度之培養基最好,另
添加生長素、活性炭、Tryptone者並不佳;Paph. philippinense以1/2MS
(A)+30g/l蔗糖濃度之培養基最好。成熟種子(Paph. purpuratum為授
粉後12個月,Paph. philippinense為授粉後4個月)經低溫(4℃)貯藏
處理,Paph. philippinense種子冷藏 2個月後播種,與未經過冷藏的種
子都在播種後兩個月內發芽,且經冷藏過的種子發芽較好;Paph.
purpuratum種子冷藏2個月後播種,有提早發芽的現象,雖比未經過冷藏
的種子遲播2個月,但卻同時發芽。

Three species of Paphiopedilum, Paph. purpuratum, Paph.
philippinense and Paph. sukhakulii were used on this study. The
development of capsules and seeds, germinating seeds to
different midia and temperatures and the effects of seedlings
to media and the seeds at 4℃ storage were investigated be
taken in this experiment. The results are summarized as follows
: 1.The seeds of Paph. purpuratum would turn into yellow 8
months after pollination. Under scanning by electron microscope
showed archesporial cells divided one month after pollination,
the megaspore developed after 2-4 months, the seed formed after
5 months, and after 11 months, the seeds developed well. The
other kind, Paph. philippinense showed 105 days after
pollination, the seeds become golden-brown color. 120 days,
dark brown, then after 110 days the capsules turned into brown.
2.The seeds were tBih- Yuan LiouHuang media and temperatures
showed Paph. purpuratum germinated best in MS medium at 25℃,
the percentage of germination was 19.5﹪, the Paph.
philippinense did best in RE medium at 25℃, the percentage of
germination was 85.5﹪. Paph. sukhakulii did the best in RE
medium at 20℃, the percentage of germination was 27.1﹪. The
protocorm development of Paph. sukhakulii showed the best
result in 1/2MS(A)+B+C+D medium at 20℃, Paph. philippinense
did the best in RE medium at 25℃, and Paph. purpuratum in MS
medium at 25℃. 3.The seedlings of Paph. sukhakulii grew best
in the medium of 1/2MS(A)+B+C+D+20g/l sugar, and Paph.
philippinense did the best in the medium 1/2MS(A)+B+C+D+30g/l
sugar. 4.The mature seeds of two species were stored 2 months
at 4 ℃, showed better germination, above all Paph. purpuratum
germinated 2 months earlier.
