以明豐、富農及屏東三批白仁萊豆種子,進行品質分析研究。富農萊豆種子重量分級處理以1.25g以上顯著提高萌芽率10%以上,泥炭土或5cm之播種深度顯著延長平均萌芽天數,於1.25g~1.75g之種子其萌芽後地上部及地下部表現均佳。大於1.50g種子,其種子萌芽力顯著高於1.25g以下之種子,而單位子葉橫切面積之萌芽力,以小於1g顯著高於其它重量之種子。
X ray條件以電壓12KV,電流2.5mA時,曝光35至55秒可獲得清晰的顯影效果。屏東萊豆種子萌芽率、株高、地下部鮮重之降低,受破裂程度與位置影響達顯著水準。胚周圍與子葉破裂顯著延長平均萌芽天數及降低萌芽整齊度。子葉破裂於胚軸周圍及破裂達1/2以上之種子顯著提高不正常苗率。浸潤速率與浸潤液導電度值之提高,於破裂嚴重種子達顯著水準。
明豐、富農及屏東種子於RH100%下經96小時後,發芽率由94%、95 %及97%分別下降為74%,83%及85%,達顯著水準,或於RH82%下經24043小時,發芽率降低為55%、67%及85%。富農及明豐老化種子平均發芽天數顯著延長約0.4~0.7天,於15℃下,顯著降低老化處理種子之發芽率,明豐、富農與屏東種子分別為31%、50%及69%,與萌芽率達顯著相關。富農、明豐及屏東種子之浸潤液導電度為96.0、115.2及85.9μmoh/cm/g,其值與萌芽率之相關係數達顯著,種子於RH82%處理10天後,導電度顯著提升為209.5、267.9及222.7μmoh/cm/g。種子TTC染色試驗中,以1、2與3染色類型之總合與萌芽率呈顯著相關。明豐種子浸潤液中全可溶性糖、胺基酸及鉀離子之滲出量均顯著較高於富農或屏東種子。依不同品質分析結果,得知三批種子品質為屏東最好,富農次之,明豐種子最差。
水化處理進行於41℃下,RH82%,72小時,提高15℃之種子發芽率26~30%與縮短平均發芽天數為1.2~1.7天,達顯著水準。水化處理降低種子浸潤液導電度值與促進種子於低溫下浸潤之呼吸率。滲調處理可提高種子發芽速度與整齊度,回乾條件以35℃顯著降低明豐種子發芽率為76.1%。於對峙培養中T. harzianum較R. solani之菌絲生長快速,經生物滲調之萊豆種子於混合介質中,播種後幼苗存活率由85%顯著提高為98.3%。

Summary
The seed quality of three seed lots of lima bean (Phaseolus liminesis Macf.), (Mean-Fan, MF), (Fu-Non, FN), and (Pin-Ton, PT) were examined. The weight of FN seeds over 1.23g significantly increased 10% emergence percentage. The mean emergence days were significantly extended when seeds were sowed in peat or in 5cm depth. The seedling growth of 1.25~1.75g seeds were better than those of the small seeds. The emergence forces per cotyledon area of 1.5~1.75g seeds were significantly higher than those of others.
12KV, 2.5mA and 35~55seconds exposure had a clear radiograph. The emergence rate, seedling height, and root fresh weight was significantly reduced by crack level and crack position. The crack around the embryo and cotyledons could extend the mean emergence days and reduce the uniform of emergence. The crack around the embryo and over 1/2 area of cotyledons increased the abnormal seedlings. The imbibition rate and the conductivity of soaking solution were increased in the seriously broken seeds.
Germination percentage of MF, FN, and PT seeds aged in RH100% for 96 hours was reduced from 94%, 95%, and 97% to 74%, 83%, and 85%, respectively, or in RH 82% for 240 hours was reduced to 55%, 67%, and 88%, respectively. The mean germination days of FN and MF seeds aged in RH 82% for 240 hours was extended for 0.4~0.7 days. At 15℃, the germination percentage of accelerated aging seeds of MF, FN, and PT was 31%, 50%, and 69%, respectively. The conductivity of soaking solution of MF, FN and PT seeds was 96.0, 112.2, and 85.9μmoh/cm/g, respectively. The correlation coefficient between the conductivity and the emergence percentage was significantly correlative. The conductivity of soaking solution of MF, FN, and PT seeds aged in RH82% for 240 hours was 209.5, 267.9, and 222.7μmoh/cm/g, respectively. The correlation coefficient between the sum of 1, 2, and 3 TTC stained pattern and the emergence percentage was significantly correlative. The total solubility sugar, free amino acid and potassium ion in soaking of MF seeds were than those of FN or PT seeds. From the result of seed quality analysis, the quality of the PT seed lot was the best, the FN seed lot was medium, and the MF seed lot was the least in three seed lots.
The germination percentage of seeds hydrated in RH82%, 41℃for 72 hours was increased 26~30%, and the mean germination days was shortened for 1.2~1.7 days at 15℃. The conductivity of soaking solution of the hydrated seed was reduced and the respiration rate of the hydrated seed at 15℃was enhanced. The mean germination day and the germination uniformity was enhanced by the solid matrix priming. The germination percentage of MF primed seed was significantly reduced to 76.1% when seeds were dehydrated at 35℃. Hypha of T.harzianum T22 growth was faster than that of R.solani in dual culture. The survival of bio-priming lima bean seedling in mixed medium was significantly increased from 85% to 98.3%.
