分子系統研究經過十餘年的發展,以DNA序列作為分子分類的方式逐漸成為潮流,除了在鑑定上,對於親緣關係、族群遺傳或演化關係的研究也多有應用。在2003年更出現了以粒線體COI基因(cytochrome oxidase subunit I)作為鑑定各類生物的論調,也就是生命條碼(DNA barcode)。本實驗以自然科學博物館冷凍遺傳物質中鱗翅目(Lepidoptera)樣本,及野外採集之天蠶蛾科(Saturniidae)樣本,建立台灣地區常見蛾類的生命條碼資料,並進一步分析其中的相關議題。共計27科147屬189種504筆序列資料完成定序,針對這些序列進行不同分類位階的變異分析。結果顯示,各科種內個體間的COI序列變異絕大多數都低於2%,僅少部份高於3%;在屬內種間的分析,發現有幾屬其種間變異低於2%,可能是因形態不易鑑定、個體變異或是近期分歧等原因所致,然而其樣本不多,尚須確認這類的情形是否普遍存在;在科內屬間的分析結果顯示,有5組屬間序列差異低於2%,經檢查後發現均屬同物異名(synonym);各科間平均變異分析結果顯示,瘤蛾科(Nolidae)與其他各科間的平均差異最小,而毒蛾科(Lymantriidae)則是最高,或許代表著毒蛾科中含有較古老的支系存留至今。加入資料庫相關序列進行分析的結果顯示,屬內種間與科內屬間變異範圍分別為4~14%及6~17%,其重疊範圍多在8~10%;在夜蛾科(Noctuidae)、蠶蛾科(Bombycidae)與天蠶蛾科內出現了不少種間變異小於2%的情形;另發現台灣的天蠶蛾與其他地區相關亞種間序列變異多數高於4%,顯示其內含有多個物種提升的可能性。本研究顯示,分子序列差異大小確實能幫助釐清不易由形態特徵發現的分類問題。

Application of DNA sequences in molecular systematics has become more prevalent in the previous decade, not only in taxonomic work, but also in many relative researches focusing on phylogeny, population genetics, and evolutionary events. In 2003, the so-called “DNA barcode” was proposed and allocated to identify organisms based on sequences of the mitochondrial COI gene (cytochrome oxidase subunit I). The study herein, DNA barcodes of Taiwan lepidopterans were established, then used to discuss the sequence divergences in each taxonomic category. The samples applying in this study were majorly derived from National Museum of Nature Science, and including some saturniids were collected from field. The result, in general, shows that the intraspecific sequence divergences are lower than 2%. However, limited cases of sequence variation lower than 2% are also found between species within a given genus and between genera within a given family respectively. Furthermore, we compiled relative sequences from NCBI database and found the sequences divergence are 4~14% and 6~17% in specific and generic categories, respectively; and the result shows the overlapping ranges of the above two categories from 7~10%. Sequences divergence in several cases less than 2% between generic species is appearing in Noctuidae, Bombycidae and Saturniidae; moreover, as comparing to other saturniid subspecies, sequences show a great divergence in subspecies collecting from Taiwan. In conclusion, barcoding analysis in this study does show the species identification possibility in additional to discover the cryptic species or refine the species resolution.
