水稻白葉枯病菌(Xanthomonas oryzae pv. oryzae, Xoo)可引起水稻白葉枯病(bacterial blight),是全世界水稻栽培區的重要病害之一。以剪葉法接種台中在來一號(Taichung Native-1, TN-1)水稻,於16天後測量其病斑長度,可得知台灣所分離的水稻白葉枯病菌菌株間致病能力差異頗大,而依其致病能力之不同,可將台灣的 Xoo 菌株區分為強、中與弱病原性菌株,其代表菌株分別為 Xoo28(強病原菌株)與 Xoo20(弱病原菌株)。為了解台灣 Xoo 菌株致病能力的差異與其基因組成之關係,利用基因減免法與南方雜合法,已找到一DNA 片段,存在於強病原菌株 Xoo28 中,但在弱病原菌株 Xoo20 中沒有發現。將 Xoo28 genomic library 得到之 BAC clone,pCHUB79 中次選殖得到的 3.5 kb之DNA差異性片段完全解序後,經 FramePlot 3.0beta 與 Vector NT1 軟體分析及 NCBI 之 Protein BLAST 比對後,發現此 DNA 差異性片段並未存在於兩個已解序之Xoo 基因體中 (Xoo KACC10331, Xoo MAFF311018) ,且只有某些台灣 Xoo 菌株有此片段,顯示此 DNA 差異性片段可能是透過水平基因轉移而得到的。

Bacterial blight of rice, caused by Xanthomonas oryzae pv. oryzae (Xoo) , is one of the important disease in many rice-growing areas. Several Xoo strains isolated from Taiwan were inoculated in rice cultivar Taichung Native-1 (TN-1), and the lesion length was measured 16 days after inoculation. According to the lesion length, the Xoo strains used in this study were categorized into severely, moderately and weakly virulent strains with representatives Xoo28 (severely virulent strains) and Xoo20 (weakly virulent strains). To understand the difference in virulence toward rice cultivar TN-1 among different Xoo strains might be due to the difference in their genome organization. Utilizing genome subtractive hybridization and Southern hybridization, a DNA fragment was identified that was present in the highly virulent strain Xoo28 and absent in the weakly virulent strain Xoo20. To facilitate DNA analysis, a BAC clone, pCHUB79, was isolated from Xoo28 genomic library and found to contain the differential 3.5 kb DNA fragment. Sequence analysis using FramePlot 3.0beta, Vector NT1 and Blast analysis showed the differential DNA fragment is not present in the two sequenced Xoo genomes (Xoo KACC10331, Xoo MAFF311018), and only some Xoo strains isolated from Taiwan contain it, suggesting the differential DNA fragment might be introduced into the Xoo strains through horizontal gene transfer.
