This study investigated supercritical carbon dioxide (SC-CO2) added with EtOH extraction to recover Scopoletin and Artemisinin from Artemisia annua. Response Surface Methodology (RSM) was applied to find operation conditions in obtaining the most weight of the extract of Scopoletin and Artemisinin and purity. The results showed that the most weight of the extract of Scopoletin at 4500psig and 40°C was 281 μg /g of feed(relative recovery was 63.37w/w %), at the same extract condition could get the highest purity but was only 0.635w/w %. The most weight of the extract of Artemisinin at 3500psig and 40°C was 11.36mg/ g of feed(relative recovery was 94.68w/w %), the highest purity at 4500psig and 60°C was 26.53w/w %。
We use normal phase Silicagel-60 resin column to purify Artemisinin that sample was extracted by add 20w/w% hexane supercritical fluid and 16-hr Soxhlet hexane. The results showed that purity is up to 98.23wt% however recovery is only 54.44 wt%.
本研究以乙醇和二氧化碳混合流體方法，萃取青蒿粉末中茛菪酯(Scopoletin)與青蒿素(Artemisinin)，並利用應答曲面法(RSM)，分析獲得兩者最大萃出量及其純度值的最適條件。實驗結果發現，當乙醇添加量(16.25wt%,105.78g)，茛菪酯的萃取條件在4500psig與40°C有最大萃出量281 μg /g of feed(相對回收率為63.37w/w %)，而在相同萃取條件下有最高純度，僅有0.635w/w %。青蒿素的萃取條件在3500psig與40°C有最大值11.36mg/ g of feed(其相對回收率為94.68w/w %)，而萃取條件在4500psig、60°C時會有最高的純度26.53w/w %。而在純化方面，本研究利用正相樹脂silica gel-60的管柱，來分離索式與超臨界流體萃出物中的青蒿素，發現可將青蒿素純度提高至98.23wt%，但回收率却僅有54.44wt%。
