摘要: 
胚珠於高等植物有性生殖中扮演十分重要的角色。我們以臺灣蘇鐵的
胚珠為材料,利用2D-PAGE的分析方法,比較蘇鐵胚珠與營養器官(根、
莖、葉),及其他生殖器官(花粉、大孢子葉、小孢子葉)之間的蛋白質
組成差異,找到5個胚珠大量表現的蛋白質及3個胚珠專一蛋白質。在三個
胚珠專一蛋白中,CTO-18是胚珠發育期間持續性表現的蛋白,而CTO-15於
胚珠發育前期表現,CTO-22則於胚珠發育的後期表現。我們針對CTO-15
及-22蛋白製備多株抗體,並以blot-affinity方法純化。Western blot結
果顯示抗CTO-15蛋白抗體除了會辨認CTO-15蛋白外,亦會辨認蘇鐵葉子蛋
白;而抗CTO-22蛋白抗體只辨認胚珠CTO-22蛋白。另外,就我們所試驗的
六種不同植物的胚珠或子房中,並無類似CTO-22蛋白的出現。CTO-22蛋白
的氨基酸序列和已知的蛋白比對後,顯示CTO-22蛋白可能為一未知的蛋白

The ovules play an important role in sexual reproduction of
higher plants. We choose ovules of Cycas taiwaniana (CTO) as
materials, and use two-dimensional polyacrylamide gel
electrophoresis (2D-PAGE) to compare the protein patterns
ofdeveloping ovules to those of other reproductive organs and
vegetative organs.Five ovule-enriched and three ovule-specific
proteins were detected during development. Of the three ovule-
specific proteins, CTO-18 was constitutively expressed during
development while CTO-15 and -22 proteins were stage-specific.
The CTO-15 protein accumulated at the early stage of ovule
development whereas the CTO-22 protein accumulated at the late
stage of development. Antisera weredirected against CTO-15 and
-22 proteins respectively, and antibodies were further affinity
purified. Western blot analysis of different organs of
cycadsrevealed that the affinity purified anti-CTO-15 antibodies
recognize CTO-15 protein and leaf proteins. On the other hand,
the affinity purified anti-CTO-22antibodies only recognize the
CTO-22 protein. No CTO-22-like protein appeared in two species
of gymnosperm and in a variety of species of angiosperm. The
partial amino acid sequence of CTO-22 protein does not show
similarity with anyknown proteins.
