先前實驗室成功的利用抑制扣除法(suppression-subtractive hybridization)建立了小孢子發育時期之鐵砲百合花藥的cDNA 集合庫,從中挑選出LLA1380選殖體作進一步的分析與探討。對此設計專一性的引子,經由5''-RACE,得到全長491 bp,編碼區有342 bp,可轉譯出113個胺基酸,預測的分子量為11.7 kDa,等電點為8.94,經由序列比對得知此蛋白與擬南芥的脂質結合蛋白有38 %的相同性。透過北方墨點法得知LLA1380在花藥發育過程中表現在花藥壁和小孢子內。進一步利用原位雜合(in situ hybridization)實驗確定LLA1380 mRNA位於花藥壁的絨氈層上。論文另一部份為LLA23蛋白質之功能分析。LLA23屬於ASR(ABA-, stress-, ripening-induced)蛋白質一員。先前的研究己知35S::LLA23在擬南芥轉殖株具耐旱和耐鹽的特性。在不同的低溫(1~5 °C)或冷凍溫度 (-2~-8 °C) 處理下,35S::LLA23轉殖株之生長均較野生型好。在零下低溫處理下(-6~-8 °C),轉殖株的存活率約高於野生型3倍;高溫(39~41℃)處理下,轉殖株的存活率也明顯較野生型好。35S::LLA23轉殖株在零下溫度的條件處理下,其離子滲漏的情況明顯低於野生型,但是溫度到-6℃以下,則差異不明顯。35S::LLA23轉殖株在高溫處理下,其離子滲漏的情況明顯低於野生型,但在低溫(1~5 °C)下與野生型沒有差異。即時定量聚合酵素鏈鎖反應的結果顯示,經由低溫和冷凍的逆境處理,百合LLA23基因表現可能在轉殖植物體內影響了相關逆境基因的表現,但對於脯胺酸的生成和ABA的合成等基因的表現,則沒有差異。
