竹子屬於禾本科中的竹亞科,竹子由於生殖性狀資訊缺乏、營養性狀易受環境影響,因此,現今越來越多的試驗由 DNA 分子研究的觀點切入,希望能克服傳統以性狀進行分類的問題。本論文選擇包含台灣特有種在內的22個竹子品種及一個水稻品種作為試驗材料,運用跨物種 SSR 應用的方式,以水稻12條染色體上平均分布的120個 SSR 分子標誌分析竹類之基因組。在獲 PCR 產物的 SSR 引子 (80/120, 66.67%)、多型性比例 (100%)、平均每個 SSR 得到的 allele 數目 (27.4) 等方面,結果皆較前人文獻中高,但不同種類的 motif 則對增幅成功率影響不大。分群分析的結果將水稻分在最外圍,以相似係數0.06把22個竹子品種分成三群,並搭配地理分布區域及地下莖生長方式,分析出22個竹子品種間之關係。

Bamboo belong to Bambusoideae, Gramineae. Information of bamboo flowering material is rare and vegetative characters are often environmentally influenced. Accordingly, more study use molecular markers to solve the problem derived from taxonomy of bamboos using vegetative characters. In this study, we used 120 rice (Oryza sativa) simple sequence repeat (SSR) markers in 22 bamboo species and one rice, O.sativa cv TNG67, as out group control. The result indicated that, 66.67% (80/120) primer pairs yielded products and were polymorphic in bamboo species, and the average number of alleles produced per primer set was 27.4. These result were better than in previous studies. But the primer amplification was not affected by different motif types. In the analysis dendrogram, rice was a outgroup and the 22 bamboo species separated into three clusters by similarity 0.06. According to geographic location and growing type (clumper or runner), these result supported the current bamboo division and provided valuable information about phylogenetic relationships.
