本試驗利用臺灣產藥用植物華薊(Cirsium lineare (Thunb.) Sch. Bip.)之莖節及葉片作為培植體,探討其大量繁殖之條件,試驗結果如下:
種子經消毒後在1/2 MS培養基中發芽之無菌苗,取單一莖節和葉片為培植體,分別接種於MS培養基添加0.2、0.5、1.0、2.0 mg/L BA與0、0.1、0.5 mg/L NAA不同濃度之組合處理,與不含生長調節劑之培養基為對照組。培養1個月後調查芽體再生及癒合組織形成結果,莖節和葉片培養皆於含有0.2 mg/L BA+0.1 mg/L NAA組合之培養基可獲得最大量增殖的芽體(莖節8.8個、葉片7.2個)。將初代培養所得之叢生芽體培養於具2-3葉片時,切取單株接種於添加0、0.1、0.5、1.0、2.0 mg/L NAA之MS培養基中誘導發根,培養1個月後,以培養於添加0.1 mg/L NAA之MS發根培養基最佳,發根率達100 %,平均根數為9.6條、平均根長4.2 cm,植株平均莖長為5.3 cm。發根後之瓶苗直接移植於泥碳土:珍珠石=1:1之栽培介質中於溫室培養,1個月後存活率為100 %,可得到生長良好之植株。

The objective of this study was to establish micropropagation system for production seedlings of medicinal plant, Cirsium lineare (Thunb.) Sch. Bip. Results are summarized as follows:
Nodal stem segments and leaf explants were taken from in vitro seedlings germinated from seeds grown in a 1/2 MS medium. Explant cultured in a basal MS medium containing with 0.2, 0.5, 1.0, 2.0 mg/L BA in combination with 0, 0.1, 0.5 mg/L NAA and with a hormone-free medium as the control in additional. The highest number of shoots (8.8 and 7.2) were obtained from 0.2 mg/L BA+0.1 mg/L NAA treatment after one month cultivation under lightness for both types of explants. The individual shoot with 2-3 leaf obtained from regeneration were subcultured in to a MS medium containing with 0.1、0.5、1,0、2.0 mg/L NAA for root formation. The best rooting(100 %)was obtained from the medium containing with 0.1 mg/L NAA after one month of culturing. The average number of roots were 9.6 with an average length of 4.2 cm from a plantlet with 5.3 cm in height.The rooted shoots were transplanted into a medium compositing with perlite and peat-moss (1:1), and an 100 % acclimation rate along with good growth for plantlets were obtained after 4 weeks of greenhouse cultivation.
