本研究由以奈和菲為基質長期馴養之化學衡定器(Chemostat)中分離出純菌,以PAHs中的雙環分子奈(Naphthalene)及三環分子菲(Phenanthrene)作為研究主化合物,進行各純化菌株的Naphthanene oxygenase比活性值的表現、生長曲線特性、奈降解效率情況、中間產物的差異、呼吸儀測試和環境因子探討,藉此比較各純化菌株以瞭解其對於降解PAHs特性和差異,期可較精確的推估土壤採生物復育的效果。
實驗將所分離出純菌株三株純菌株以16S rRNA進行鑑定,其結果分別為Comamonas testosteroni、Rhodococcus ruber、Serratia marcescens。從實驗求得三株純化馴化菌的Km、Vmax 、Y、k值和 k/Y值。對於Naphthanene oxygenase動力學參數,Km值分別為0.26、0.19和0.24 mmole;Vmax值分別為0.00047、0.00084和0.00056 mmole/min。對於各菌株降解奈的Y值分別為0.77、0.63和0.87;k值分別為0.35、和0.29和0.31 hr-1以及k/Y值分別為0.45,0.47和0.35 hr-1。在呼吸儀測試中對於各菌株的總耗氧量和理論耗氧量比值,在以奈(溶解態)為基質時分別為0.77、0.66、和0.91;奈顆粒(結晶狀態)時為基質分別為0.29、0.97和0.87和以菲為基質時分別為0.01、0.08和0.02,實驗結果可知三株菌能將奈有效的礦化而對於菲卻不行。環境因子對各純化菌株影響實驗,得知各菌株於微酸性(pH=6.5)水相環境中、較高溫度(34℃)且氮源為銨態氮的型態時較有利於生長。環境中氧氣的供應量對於Comamonas testosteroni是非常的敏感,而對於Rhodococcus ruber則無明顯之影響性。。

ABSTRACT
The objective of this study was to assess the biodegradation of polycyclic aromatic hydrocarbons (PAHs) by the acclimated bacteria. This study focused on the rate of biodegradation、the growth rate、the specific enzyme activity、the oxygen uptake rate and environmental factors for PAHs biodegradation.
Three acclimated PAHs-degrading strains,Comamonas testosteroni,Rhodococcus rubber and Serratia marcescenswere,were isolated from a chemostat,which was continuously fed with naphthalene and phenanthrene as the substrates. In the specific enzyme activity tests,the experimental results indicated that the value of Km were 0.26、0.19 and 0.24 μmole;the value of Vmax were 0.00047、0.00084 and 0.00056 μmole/min for these three species,respectively. In the biodegradation rate and growth rate tests,experimental results indicated that the value of Y were 0.77、0.63 and 0.87;the value of k were 0.35、0.29 and 0.31 hr-1 and the value of k/Y were 0.45,0.47 and 0.35 hr-1. for these three species respectively. The ratios of the theoretical oxygen demand to the experimental oxygen demand obtained from a respirometer with soluble NAP as the substrates were 0.77、0.66、and 0.91 for these three species,respectively. However the ratios were 0.29、0.97 and 0.87 for these three species,respectively when coystal NAP was the substrate.When phenanthrene was used as the substrates,the ratios were 0.01、0.08 and 0.02 for these three species,respectively. For environmental factors stady,the experimental results indicated that these three acclimated degrader grew batter in the temperature at 34℃ and pH at 6.4,especially using ammonium as the nitrogen sources.
