標題: 國產基因改造木瓜回交商用果實之輪點病毒鞘蛋白基因在動物體內重要器官表現、免疫毒理及對大鼠90天餵食試驗之安全性評估

本實驗室於99及100年度為配合國內已進入田間試驗之基因改造木瓜,已完成評估針對木瓜輪點病毒單抗18-2-4與單抗親本非基改台農2號回交後(編號22-10)及單抗親本非基改泰國株木瓜回交後(編號823)及回交後編號22-10與823雜交商業品種F1果實之基因毒理及動物毒理試驗,結果顯示國產基因改造木瓜回交果實各個品種於基因毒理試驗中皆不具致變異之毒性作用,且對大鼠口服急毒性及連續餵食28天動物毒理試驗亦無明顯毒性反應。本計畫參考歐盟及國內「階段性測試體系(Hierarchical Test System)」第三階段之「基因改造食品安全性評估毒理參考手冊」之建議,進一步進行PRSV基因改造木瓜基因片段在體內分佈、免疫毒理及90天餵食毒性試驗,以提供更完整安全評估之參考。擬分2年完成,第1年擬進行回交商業品種第一代(F1)果實(823-2210)基因改造木瓜經餵食大鼠後,分析其體內重要器官,如腦、心臟、肝臟、脾臟、肺臟、腎臟、睪丸、胃腸內容物、尿液及血液之木瓜內源性基因片段papa32及PRSV基因改造木瓜基因片段之存在與否及其體內分佈。另經餵食小鼠後之免疫毒理及特異性免疫蛋白表現,分析其體內脾臟及週邊血液淋巴細胞之T 細胞(CD3+)及B細胞 (CD19+)細胞族群分佈,CD4+, CD8+及炎症相關之細胞激素IL-2, 4, 5, 10, 12, TNF-, IFN-, MCP-1 (monocyte chemoattractant protein-1)及MIP-1 (macrophage inflammatory protein-1)調控變化,且進行免疫球蛋白(IgG, IgM, IgE, IgA)含量之分析,以了解食用商用基因改造木瓜對體內免疫毒理之安全性。第2年進行回交商業品種第一代(F1)果實(823-2210)基因改造木瓜90天餵食毒性試驗。

Last two years, we have performed to evaluate the food safety of the newly development of backcross genetically modified papaya fruits in Taiwan. The backcross fruits, including mono-resistant transgenic papaya lines (18-2-4) protected against Papaya ringspot virus backcrossed with the non-GM control line of Sunrise (code no. 2210), or non-GM control line of Thailand (code no. 823) and the other one was hybrid with 2210 and 823 (code no. 823-2210). Results revealed that whole foods of genetic modified plant foods have no genetic and animal toxicities between the parent fruits on these studies. We reviewed the approaches of genetic modified plant foods based on the FAO/WHO recommended guidelines and literature references for genetic modified foods that has published by the Department of Health, the European Commission (EC), USA and other countries related GM regulations. The proposal of toxicological assessment approach is to meet on the third step of Hierarchical Test System for genetic modified plant foods evaluation. The objective of this project will aim to evaluate the gene expressions in body, immuotoxicity and long-term feeding toxicity of commercial line of hybrid fruits (code no. 823-2210) in more detail. It will be taken two years, the first year is to determine the gene expressions of endogenous papa32 and PRSV in the brain, heart, liver, lung, spleen, kidney, testis, stomach, intestine, urine and blood, and the immune responses of lymphocytic activities including CD4+, CD8+ in the spleen and peripheral blood and the cytokine-related modulation of IL-2, 4, 5, 10, 12, TNF-, IFN-, MCP-1 (monocyte chemoattractant protein-1), MIP-1 (macrophage inflammatory protein-1) and immunoglobins of IgG, IgM, IgE, IgA in mice. The second year will perform the 90-day feeding toxicity of commercial line of hybrid fruits (code no. 823-2210) in rats. These studies will provide useful safety evaluation for hybrid of genetic modified plant foods and promotes the production and development of other genetic modified plant foods in Taiwan.
