本研究之目的是以管柱模擬受四氯乙烯污染的地表下飽和含水層,植入具有脫氯能力之脫鹵混合菌(厭氧混合菌及馴化混合菌),進行生物復育,探討植種菌於模擬含水層中的傳輸與消長,以及對四氯乙烯的降解效應。
研究結果顯示生物刺激階段,模擬含水層中現地菌皆能忍受四氯乙烯的毒性,但無脫氯的能力。而生物添加階段,首先植種厭氧混合菌至模擬含水層中,僅能利用產甲烷之共代謝機制將四氯乙烯脫氯至三氯乙烯,且甲烷菌無附著生長於模擬含水層中;接著植種馴化混合菌至模擬含水層中,可將四氯乙烯脫氯至順-二氯乙烯,且操作 16 天後之四氯乙烯去除率約有 90 %,而 65 天後四氯乙烯之去除率高達 99 %,但此階段之脫氯作用主要受限於順-二氯乙烯的生物分解。
批次試驗結果顯示,管柱中菌液於 31 天內僅能將四氯乙烯脫氯降解至順-二氯乙烯,而馴化混合菌於四氯乙烯降解過程中,無氯乙烯的累積即有乙烯的生成,且四氯乙烯之生物降解皆屬於一階動力反應。
馴化混合菌植種至模擬含水層中,可觀察到 Clostridium lactatifermentans 與Dechloromonas sp. EMB 50 隨著水流方向傳輸,且皆於 day 35 後遍佈於模擬含水層中,表示基質與營養鹽供應是適當的,以避免短時間內大量微生物生長形成阻塞而降低生物復育的成效。此外,經由螢光原位雜交(FISH)結果顯示,馴化混合菌中具有 Dehalococcoides spp.,植種至模擬含水層後 65 天遍佈於其中,且數量隨流經距離的延長而有降低的趨勢。

The objective of this study was to explore the growth and distribution of mixed culture with dechlorinating capability in a sand column simulating a PCE-contaminated aquifer.
The results of biostimulation study indicated that the indigenous in the simulated saturated aquifer could tolerate the toxicity of PCE, but without dechlorination capability. With the bioaugmentation of anaerobic mixed culture, PCE could be biotransformed to TCE. However, with the bioaugmentation of PCE-acclimated culture, PCE could be biotransformed to cDCE. After 16 and 65 days of operation, PCE removal could be up to 90 % and 99%, respectively. But cDCE could not be further de dechlorinated.
In microcosm study, PCE could be dechlorinated to cDCE after 31 days of operation. Ethene could be observed in this column study. However, VC was not observed. Biodegradation of PCE followed first-order reaction.
With the addition of PCE-acclimated culture in this saturated aquifer, Clostridium lactatifermentans and Dechloromonas sp. EMB 50 distributed along the column after 35 days of operation. Although Dehalococcoides spp. was presence in the column, its concentration decreased along the column.
