標題: 乳酸菌菌體及胞內物抗氧化性之探討

摘要: 
乳酸菌抗氧化性試驗主要是以抑制liposome過氧化、細胞脂質過氧化、清除DPPH、抑制deoxyribose降解、comet assay及MTT細胞存活率分析進行探討,分析106-109 cfu/ml的乳酸菌菌體及胞內物之抗氧化活性。結果顯示在liposome的系統中,以S. thermophilus具有最好的抑制liposome過氧化物產生的效果,尤其又以Streptococcus thermophilus 821的抗氧化活性最高,於109cfu/ml之菌體及胞內物可高達76%及56﹪的抑制效果。於抑制細胞脂質過氧化方面,以L. bulgaricus Lb對MDA的生成有極佳的抑制效果,於109 cfu/ml之菌體及胞內物可使MDA的生成量由4.8μM降至1.4 及1.3μM。而在清除DPPH系統中則以Lactobacillus bulgaricus 1006的清除能力最強,菌體於109cfu/ml可高達87.5%的抑制效果。在評估DNA傷害方面,以B. longum此菌的保護效果遠優於其他三菌,又以B. longum 15708這株菌的抗DNA傷害能力最好。同樣地,去氧核醣降解試驗也為評估DNA傷害試驗,發現以B. longum B6的效果最好,於109 cfu/ml可高達88.6%的抑制率。而在探討乳酸菌對抑制H2O2所引起的細胞毒性方面,則採用MTT細胞存活率分析,結果以S. thermophilus 3641降低細胞毒性的效果最好,於109 cfu/ml菌體及胞內物的細胞毒性抑制率高達87%及70﹪。上述試驗中當菌數提高時,抗氧化活性也隨之提高,且大部分的菌株在108-109 cfu/ml的菌數時可達到顯著的抗氧化活性。

The experiments of 106、107、108 and 109 cfu/ml of intact cells and intracellular extract of lactic acid bacteria antioxidative activity including lipid peroxidation of liposome、scavenging DPPH、deoxyribose degrandation、MTT cell viability assay、lipid peroxidation of cell and comet assay were studied. Among 19 strains, Streptococcus thermophilus 821 reveals the best inhibitory effect of lipid peroxidation of liposome at 76% of 109 viable lactic acid bacteria. L. bulgaricus Lb shows the best inhibitory effect to reduce MDA concentration of Intestine 407cell line at 1.39 µM of 109 viable lactic acid bacteria. Lactobacillus bulgaricus 1006 had highest scavenging DPPH radical ability at 87.5% scavenging effect of 109 viable lactic acid bacteria. In the aspect of comet assay, Bifidobacterium longum 15708 shows the best protective effect of DNA from H2O2 injury among 19 strains. B. longum B6 had the best effect to inhibit deoxyribose degrandation at 88.6% inhibitory effect of 109 viable lactic acid bacteria. S. thermophilus 3641 revealed the best inhition of cytotoxicity of Intestine 407 at 87.4% of 109 viable lactic acid bacteria. Above experiments, the cell number of lactic acid bacteria is increased the antioxidative ability of lactic acid bacteria increased. Most strains showed significant antioxidative activity at 108-9 viable lactic acid bacteria. Moreover, antioxidative ability of intact cells is better than intracellular extract.
