本實驗以19株乳酸菌包括Bifidobacterium longum 15708、B6、Lactobacillus acidophilus 4356、B、E、Farr、LA-1、N-1、Lactobacillus bulgaricus 448、449、1006、11842、12278、Lb及S. thermophilus 573、821、3641、19987、MC,來進行對酸與膽鹽oxgall之耐受性、於含不同類型膽鹽之培養基下降膽固醇之情形、膽鹽水解以及對結合型膽鹽行去結合能力的測試。根據實驗的結果,19株乳酸菌皆可以在pH 3下存活至少3小時,B. longum 15708、L. acidophilus 4356、N-1、L. bulgaricus 449及S. thermophlius 573、3641對酸性的耐受性較高,能在pH值2下超過2小時;當乳酸菌培養於含膽鹽oxgall之培養基中時,顯示S. thermophilus 573具有最高之降膽固醇作用,可降低約42%之膽固醇,其次為L. acidophilus 4356的36%;而L. bulgaricus 12278、L. acidophilus 4356則分別在含有膽鹽cholic acid及taurocholic acid的培養基中具有最大降膽固醇的能力,分別為25%及24%。其次,本研究進行膽固醇於酸性pH值下與不同類型膽鹽培養基之測試,發現膽固醇與去結合型膽鹽發生共同沉澱。B. longum 15708、L. acidophilus 4356及Farr同時具有對taurocholic acid及taurodeoxycholic acid之膽鹽水解活性。對酸及膽鹽耐受性的菌株,對膽固醇的移除有正面之影響,但與乳酸菌是否具有膽鹽水解並無直接之關聯;而乳酸菌之膽鹽水解活性之大小,也對其降膽固醇的能力影響不大,因膽鹽水解活性大小,只影響膽固醇與去結合膽鹽共同沉澱的程度。因此,乳酸菌之降低膽固醇的機制包括膽鹽水解之去結合作用,以及膽固醇與去結合型膽鹽於酸性pH值下所產生共同沉澱與乳酸菌細胞的作用。

Nineteen strains of lactic acid bacteria including Bifidobacterium longum 15708、B6、Lactobacillus acidophilus 4356、B、E、Farr、LA-1、N-1、Lactobacillus bulgaricus 448、449、1006、11842、12278 and Streptococcus thermophilus 573、821、3641、19987、MC were tested acid and oxgall tolerance、cholesterol reducing ability、bile salt hydrolase activity and deconjugation ability. Most of the strains survive for 3 hours in pH 3. B. longum 15708、L. acidophlus 4356、N-1、L. bulgaricus 449 and S. thermophilus 573、3641 can survive more than 2 hours in pH 2; S. thermophilus 573 reduced cholesterol about 42%, and L. acidophlus 4356 reduced cholesterol at 36%. L. bulgaricus 12278 and L. acidophlus 4356 reduced cholesterol at 25% and 24% while strains incubated in broth containing cholic and taurocholic acid. Also the tests were found cholesterol copreceptated with deconjugated bile salt when different type of bile salts in acidic pH. B. longum 15708、L. acidophlus 4356 and Farr have taurocholic and taurodeoxycholic acid bile salt hydrolase. Acidic and bile tolerance can help strains to reduce cholesterol, but there is no relation with bile salt hydrolase. The level of bile salt hydrolase can not influence the reduction of cholesterol. It only change the degree of the copreceptation of cholesterol and deconjugated bile salt. The mechinism of reduction of cholesterol including deconjugation、assimilation and copreceptation of cholesterol and deconjugated bile salts.
