dc.description.abstract | 苯丙胺酸(Phenylalanine)是人體所須之必需胺 基酸,近年來由於人工甜味料Aspartame的發展成 功,使得其前驅體-苯丙胺酸之需求量激增,預估 1992年的需求量可達8,800公噸,其發展未可限量. 苯丙胺酸可以用發酵法或酵素轉換法生產,惟 酵素轉換法之成本較高.目前世界各國已趨於 利用價格低廉之糖質原料發酵生產苯丙胺酸, 所使用的菌株多為傳統的營養要求性或代謝調 節性變異株.最近,新的基因操作技術如細胞融 合、基因重組,亦被應用於改良苯丙胺酸生產 菌.主持人已發展成功E. coli coryneform bacteria之 穿梭載體,並建立利用電轉形法,高效率將載體 轉形入Coryneform bacteria.利用個人建立之基因選殖系統,已選殖出與苯丙胺酸生合成最具關鍵 性之3-deoxy-D-arabino-heptulosonate-7-phosphate synthase 基因(簡稱DAHPsynthase基因).目前正在探討DAHP synthase之回饋抑制作用位置.本計畫則在於分析 Prephenate dehydratase的回饋抑制作用位置,以便將 來能利用人為的方法,完全解除DAHP synthase及 Prephenatedehydratase的回饋抑制反應,以提昇菌體 對苯丙胺酸之產率(Productivity)及收率(Yield).本 年度的目標如下:(1)分離對m-flurophenylalanine(mFP, 屬於Phenylalanine analog)具有不同抗性強度的變異 株,並確定mFP的作用位置是在Prephenate dehydratase; (2)利用PCR方法選殖mFP-resistant prephenate dehydratase 基因;(3)mFP-resistantprephenate dehydratase的DNA sequencing;(4)比較mFP-sensitive和mFP-resistantprephenate dehydratase的Peptide sequence,以尋找 Prephenate dehydratase被Phenylalanine回饋抑制的可能 位置. | zh_TW |
