標題: 利用合成Peptide研發禽流感H5血清亞型抗體ELISA診斷套組(產學合作)

Hemagglutination inhibition (HI) test is the traditional way to determine the H subtypes of avian influenza antibodies, however, the use of viruses in the test is time consume and harzardous. In our laboratory, we developed E.Coli expression system to express the specific H gene fragment for each IV H subtype. Application of the expressed recombrinant peptide in Western blot and ELISA can specifically identify the H subtype antibody. Again it's time consume in Western blot assay, and there were high background reading in ELISA. In this proposal, we try to establish aspecific ELISA Kit for high pathogenic avian influenza H5 antibody surveillance.

家禽流行性感冒抗體血清H亞型之鑑定以往用HI test需要使用15種H亞型病毒,費時且有病毒流出之危險,本實驗室研發了用大腸菌表現15種亞型H基因片段的重組蛋白,用SDS-PAGE與ELISA可區別抗體H亞型,然而仍有費時與背景質偏高的缺點,因此本計畫擬以人工合成各H抗原特定胺基酸序列之蛋白建立簡便特異之ELISA Kit,作為高病原性禽流感H5亞型抗體監控.
