摘要: 
For developing non-pest area of export orchid, the project will investigate the flora of microbial pest in orchid garden and cultural media continually. Furthermore, good protecting material will be selected for controlling the population of microbial pest. In fungi, three objectives will be carried out, including (1) collection and identification of fungal isolates from export orchid garden, (2) examining the relationship between temperature and development of fungal diseases on orchid, (3) selecting the highly effective and lowly toxic fungicides for decreasing the population of fungi what may ause diseases in orchid, and (4) evaluation of effect of fungicides to growth of orchid. In bacteria, the Standard Operating Procedure (SOP) will be developed and examined, including (1) examining the specific and accuracy of PCR-based technology in bacterial diseases of phalaenopsis, (2) examining the accuracy of multiplex-PCR-based technology in bacterial diseases of phalaenopsis, (3) establishing data of characteristics of typical symptom and colony of phalaenopsis bacterial pathogen, (4) developing the SOP and process training, (5) application of SOP in different bacterial diseases in phalaenopsis, (6) giving assistance for bacteria identification in phalaenopsis.

為建立外銷蘭花之非疫區,本計畫將延續97年計畫,調查外銷蘭園區內及介質所帶的有害微生物相,並篩選具降低可能為害蘭花植物之有害微生物族群密度的合適藥劑。有害真菌方面,包括(1)持續蒐集及鑑定外銷蘭園內所蒐集之所有真菌;(2)參考外銷運輸期間之環境條件,以孢子或菌絲進行接種實驗,確定可能病原之發病情形;(3)篩選廣效性低毒性之殺菌劑,降低園區內可疑病原真菌的密度;(4)評估有效藥劑對蘭花生長及開花之影響。而有害細菌方面,擬建立標準檢測流程,包括(1)測試已開發蝴蝶蘭細菌性病原之專一性PCR檢測技術在田間樣品之再現性;(2)測試可同時檢測三種蝴蝶蘭細菌性病原之多引子組PCR技術在田間樣品之準確度;(3)彙整蝴蝶蘭細菌性病之典型病徵及其菌落型態;(4)建置蝴蝶蘭細菌性病原標準檢測流程,可讓檢測人員瞭解檢測流程;(5)建立標準檢測流程執行多種細菌性病害之檢測;(6)協助防檢局鑑定外銷蝴蝶蘭之細菌性病原及相關事項。
