摘要: 
本訂畫擬針對台灣分離而得的十字花科黑腐病菌建立選擇性增量流程,同時製備抗十字花科黑腐病菌表面蛋白之血清及建立專一性 PCR 檢測技術,未來將可利用選擇性增量及免疫磁珠吸附菌體的方法提高偵測十字花科黑腐病菌的靈敏度。本訂畫另針對疑似病毒感染所引起十字花科蔬菜嵌紋輪紋及enation等病徵進行診斷鑑定。工作項目包括病毒之分離與純化、病毒抗血清之製備與血清學試驗、病毒外鞘蛋白基因之選殖與定序等,預期將可產出專一性抗血清、分子探針與引子對,並發展鑑定與檢測技術,供田間及苗圃檢測之用。

The project will focus on (1) developing standard operation procedures for specific and sensitive detection of Taiwan-isolated Xanthomonas campestris pv. campestris from crucifer seeds using unique selective enrichment method and immunomagnetic separation polymerase chain reaction; (2) the isolation and purification of the causal agent of crucifer plants showing symptoms of mosaic, ringspots and enamtion, and developing specific detection tools, such as antiserum and molecular probes for precise and reliable identification of the virus.
