dc.description.abstract目前國外發展出抗性除草劑轉殖作物中,其抗性基因包括目標酵素蛋白(targetenzyme protein),或是代謝除草劑之酵素蛋白之編碼基因,均來自微生物,包括E. coli.及土壤中的細菌等。顯然於土壤中所含有的豐富微生物相(flora)提供了抗性基因之重要來源。本研究根據2005 本研究室與興大土環系楊秋忠教授合作所執行之農委會研究計畫成果,從中選出四株菌種,包括Burkholderia sacchari、Pseudomonascitronellolis、Pseudomonas psychrotolerans 與Serratia marcescens,其中B.sacchari 與P. psychrotolerans 均具高度降解能力,而B. sacchari 與S.marcescens 在高濃度glufosinate 處理下,其生長不受影響,明顯對glufosinate具抗性,唯S. marcescens 屬於腸內菌。本研究擬以P. citronellolis 為對照組,探討菌類耐受glufosinate 及其降解glufosinate 之特性,擬針對影響耐性(抗性)之targetenzyme-GS (EC 6. 3. 1. 2)之酵素反應進行酵素動力學分析,以確定其差異性。其次,擬針對代謝glufosinate 之相關酵素蛋白進行分析,由其代謝產物與相關代謝酵素活性變化,比較菌種間之差異性,確定其降解能力高之原因,以利尋獲關鍵酵素蛋白及其基因,運用於抗性轉殖作物或利用於清除環境中殘留之除草劑。zh_TW
