dc.description.abstract高粱植株以孕穗期浸水最敏感,孕穗期浸水三 至五天幼穗變小,粒數減少,產量驟減,分蘗側出. 本年度主要了解高粱在孕穗期浸水產量減少的原 因.選用雜交高粱台中五號為試驗材料,盆栽及田間 栽培於高粱孕穗期進行五天的淹水處理,分期取 樣觀察幼穗分化發育變化,並取樣1g鮮重,加入PCA 均質研磨,離心後取上層液注入HPLC測定PAs含量,另 外測定乙烯含量,以探討其間關係;此外人工淹水 濕害後外加施用PAs以了解恢復傷害的效果.孕穗期浸水造成產量減少,主要是因小穗粒數減 少,及粒重之減少,本年度計畫從乾物質生產分配 方面來討論產量減少原因.以CO/sub 2/紅外線分析 儀測定植株光合作用變化,並且間隔取樣分析各 部位乾物質分配變化,主要酵素PEPcase活性變化和氣孔阻力大小,及葉片水分潛勢的變化.PEPcase測定 依Nakamoto等(1983)分析方法,稱鮮重約0.2g,加入萃取 液磨至漿狀,離心後取上層液再加PEPcase試劑,在 Spectrophotometer 340nm下測定單位時間內的吸光度.zh_TW
