The Antioxidant and Prooxidant of Beta-Carotene and Lycopene in Human Skin Fibroblasts under Oxidative Stress

近年來由於服用β-carotene補充劑的人體試驗發現:補充高劑量的β-carotene不僅沒有降低肺癌發生率的效果,甚至會增加抽煙者及石棉工廠工作者的肺癌發生率,因此β-carotene 的抗氧化性及效果深受質疑。而另方面,lycopene的抗氧化及抗癌性卻日漸受到重視,但其效果是否優於β-carotene,尚有許多問題有待釐清。因此本實驗的主要目是要比較在不同的氧化條件下,lycopene與β-carotene的抗氧化性或促氧化性。 以人類皮膚纖維母細胞( Human foreskin fibroblast; Hs68 )之細胞培養模式,在細胞併入β-carotene及lycopene後,分別以Fe/NTA、AAPH、AMVN誘發氧化反應,探討不同的氧化壓力下,細胞膜上之�-carotene及lycopene對細胞型態、細胞膜脂質過氧化及DNA氧化傷害之影響。結果發現以Fe/NTA誘發氧化反應時,lycopene與β-carotene皆能抑制脂質過氧化,但卻促進AMVN誘發的脂質過氧化反應,而對AAPH誘發的氧化反應效果則不明顯。氧化劑AMVN對兩種類胡蘿蔔素的損耗則明顯快於另外兩種水溶性氧化劑。DNA傷害方面,除Fe/NTA處理,富含兩種類胡蘿蔔素組細胞8-OH-dG含量略小於控制組外, 其餘結果與細胞型態改變方面皆顯示,不論β-carotene或lycopene均不具保護效果,甚至有促進氧化傷害之傾向。因此Lycopene 像�-carotene般,在某些條件下可能作為促氧化劑。 The effects of �-carotene and lycopene on lipid peroxidation, DNA damage, and the change on morphology of human foreskin fibroblast cells (Hs68) induced by Fe/NTA, AAPH and AMVN individual were examined. The results show that �-carotene and lycopene act as antioxidants on lipid peroxidation induced by Fe/NTA, as prooxidants when lipid peroxidation induced by AMNM. There is no effect on AAPH-induced lipid peroxidation. The rate of �-carotene consumed in cell membrane induced by AMVN is faster than AAPH and Fe/NTA. No protection on DNA damage and the change on morphology was observed at present of �-carotene and lycopene except the Fe/NTA-induced 8-OH-dG formation. Thus lycopene act as a prooxidant in some conditions.

The interventional trials snowed that -carotene supplimentation reduces risk of lung cancer; in fact, increased risk was observed in smokers. Therefore, that whether -carotene is a efficient antioxidant is of concern. On the other hand, there were more and more attention on the antioxidant and anticasinogenic activity of lycopene. Is lycopene a better antioxidant than -carotene that need more studies. The aim of this study is to compare -carotene and lycopene the antioxidant and prooxidant activities.
