Some difficult proteins must be expressed by inducible system to avoid the modification on DNA sequences or degraded by host protease. Lactococcus lactis is a food grade host and Bacillus subtilis ia a GRAS host. In this project, the strong constitutive expression signals which were constructed previously will be developed into inducible systems。These systems will be constructed into food grade inducible systems by using food grade selection marker nisin. Difficult proteins - recombinant sweet protein brazzein and intestinal damage repairing protein, recombinant trefoil factor 1 (TFF1) will be produced by these food grade/GRAS grade inducible systems. This project aims to improve the production level of local food biotechnology industry. |
