The present study was to investigate the speculation and optimize the production medium for pigment production by Monascus pilosus ATCC 22080 in shake-flask experiments. The medium containing 2% maltose and 0.1% peptone was found to aid its sporulation. In the work, pre-cultures was used to induce mycelium growth, which was then transferred to produce Monascus-pigments. The Plackett-Burma method and two-level factorial designs were used to screen the important ingredients, followed by the method of path of the steepest ascent and response surface methodology. Using a 3-factor-5-level central composite design, the system response was optimized at: Glycerol, 32.9; Maltos, 84.4; KH2PO4, 13.8; glucose, 3; MSG, 12.5; NH4NO3, 1.5. The process for Monascus-pigment production was thus significantly shortened. Such experiments described that maltose was the best carbon source, and a high concentration of maltose greatly enhanced product formation.
本研究以紅麴菌Monascus pilosus ATCC 22080為紅麴色素生產菌株，碳源豐富而氮源限制有助於此菌株之產孢，實驗中並以搖瓶實驗探討生產培養基之最適化。本文是接種前培養菌絲來生產天然色素，實驗上利用Plackett-Burman實驗設計及兩水準部分因子設計法先篩選出重要的組成分，配合陡昇路徑與回應曲面法尋求極值點，3因子5水準的中心混成設計之實驗結果顯示最適化生產培養基為(g/L)：Glycerol，32.9；Maltose，84.4；KH2PO4，13.8；Gluscose，3；MSG，12.5；與NH4NO3，1.5；相較最適化之前，紅麴色素之生產由本研究初之十天大幅縮短為四天；實驗數據說明Maltose為最佳碳源，也說明了高濃度Maltose有助於紅麴色素之生產。
