本研究主要目的在進行半夏(Pinellia ternata (Thunb.) Breit. 之基源鑑定、品質成分分析及優良農業操作(GAP)模式研究。
將半夏以植物型態與植物組織切片法與成分指紋圖譜進行基原鑑定分析比對。結果顯示,半夏植株地上部與虎掌半夏(Pinellia pedatisecta)、水半夏(Typhonium flagelliforme)、犁實尖半夏(Typhonium divaricatum.)外觀形態上有明顯差異,辨識正品半夏基原必須以組織切片法觀察其芽痕位置、澱粉粒薄壁細胞分布、有無分生組織辨別。正品半夏與虎掌半夏都屬芽痕位於塊莖上部中央,鬚根痕集中於芽痕周圍或上部;而水半夏和犁實尖半夏屬芽痕多偏向一側,鬚根痕稀疏遍布塊莖全體。半夏其木栓層附近極少或沒有或內層均有含澱粉粒薄壁細胞,而虎掌半夏、水半夏、犁實尖半夏則均勻分布在薄壁細胞。從橫切面邊緣觀察,半夏並無分生組織,而虎掌半夏、水半夏、犁實尖半夏均有分生組織。
半夏組織培養結果顯示,將半夏接種於1/2MS+1 mg/l BA培養基中,培養6-7週即可完成1個繼代世代。因此,未來可將此技術應用在大量繁殖無病毒之半夏種苗上。
半夏肥料試驗試驗結果顯示,混合施用有機肥4000 Kg/ha與鉀肥50 kg/ha可以獲得較高的塊莖產量。進一步以HPLC分析台灣半夏品質,結果顯示台灣與中國四川半夏的Adenine含量相近,以甘肅半夏Adenine含量最少。利用多項式迴歸分析台灣半夏不同大小塊莖之成分,以半夏塊莖約3~3.4 g之指標成分最高,品質最佳。
