摘要: 
The methanol extract of the aerial part of Portulaca oleracea Linn wasdissolved into water and partitioned successively with n-hexane andn-butanol. In this research,we focus on the bioactive compounds in then-hexane、n-butanol and water soluble fractions.

The n-hexane、n-butanol and water fractions were separated by columnchromatography on silica gel,gel filtration (Sephadex LH-20),reverse phaseODS column and macroporous resin chromatography (Diaion HP-20) to givenine compounds. Three compounds were isolated in n-hexane fraction,theother one was sterols: β-sitosterol(PH-1),two of those were triterpennoids:lupeol(PH-2)、friedelin(PH-3). One compound was isolated from water solublefraction,the one was a lignan:5,5'-dimethoxy-7,7'-dihydroxy-9'-β-glucopyranosyloxyl secoisolariciresinol(PW5). Another five compounds were isolated in n-butanol soluble fraction,there were two flavonoid C-glucosides including:6-C-α-arabinopyranosyl-8-C-β-glucopyranoside(PB-4),Apigenin 6-C-β-xylopyranosyl-8-C-α -arabinopyranoside (PB-5),one phenol:ferulic acid(PB-6),two of those were flavonoid O-glucosides:Kaempferol3-O-β-D-glucopyranoside(PB-7), Quercetin 3-O-β-D-glucopyranoside(PB-8),Among them of one compound PW5 has not been reported from literature.

In addition, we studied the qualitative analysis of the six compounds inn-butanol and water fractions of aerial part of Portulaca oleracea Linn byHPLC/UV. On the chromatogram, it showed six peaks in accordance with thefingerprint of the methanol extract. Their retention times were as follow:16.2min (PW5)、17.4 min (PB4) 20.2 min (PB5)、、22.2 min (PB6) 26.1 min (PB7)、、30.5 min (PB8).

Finally, the biological activity displayed that among the above compounds,PH1 and PH2 showed inhibition on cell growth of renal cancer cells (786-O)and rat kidney fibroblasts (NRK-49f). The values of 50% cytotoxicconcentration (IC50) of PH1 are 301 and 282 μM, respectively. The values ofIC50 of renal cancer cells (786-O) ofPH2 are 267 μM

本實驗將馬齒莧(Portulaca oleracea Linn)地上部分以甲醇浸泡萃取 過,濾後經減壓濃縮得到的甲醇粗萃物溶於水,分別與正己烷、正丁醇做分配萃取,得到各粗分配層。本實驗進行水可溶層、正己烷可溶層與正丁醇可溶層成分之單離與鑑定。

利用矽膠管柱層析法、膠體過濾法(Sephadex LH-20)、大孔吸附樹脂層析法(Diaion HP-20) 及逆相 ODS 填充管柱等進行分離純化共分得 9 個化合物,於正己烷層中分離得到 3 個化合物,分別為 1 個固醇類(Sterol):β-sitosterol(PH-1),2 個三萜類(Triterpennoid): lupeol(PH-2)、friedelin(PH-3) 於水可溶層中分離得到 1 個化合物 為 1 個木酚素類(lignan):5,5'-dimethoxy-7,7'-dihydroxy-9'-β-glucopyranosyloxyl secoisolariciresinol(PW-5)。 於正丁醇可溶層中分離得到 5 個化合物,2 個黃酮碳苷類(flavonoid C-glucosides):Apigenin6-C-α-arabinopyranosyl-8-C-β-glucopyranoside(PB-4),Apigenin 6-C-β-xylopyranosyl-8-C-α -arabinopyranoside (PB-5), 1 個酚類(phenol): ferulicacid (PB-6),2 個黃酮氧苷類(flavonoid O-glucosides):Kaempferol3-O-β-D-glucopyranoside(PB-7), Quercetin 3-O-β-D-glucopyranoside(PB-8),而其中化合物 PW5 為文獻中不曾報導之新化合物。

利用高效能液相層析儀進行定性分析,將馬齒莧地上部分萃取物之指紋圖譜與分離所得的六個標準品進行比對,其滯留時間分別為 16.2 min(PW5)、17.4 min (PB4)、20.2 min (PB5)、22.2 min (PB6)、26.1 min (PB7)、30.5 min (PB8)。在成分之活性測試中,結果顯示 PH1 和 PH2 對於人類腎臟癌細胞株(786-O)和老鼠腎纖維母細胞(NRK-49f)具有抑制效果 其致死之 IC50 數值:786-O 細胞分別為 301μM、267μM,PH1 對於 NRK-49f 細胞分別為 282μM。
